高效液相色谱法测定止痒消炎水中蛇床子素含量

摘　　要：目的:建立高效液相色谱(HPLC)法测定止痒消炎水中蛇床子素含量的方法。方法:采用HPLC法,以蛇床子素为目标成分,色谱柱为ZORBAX Eclipse Plus C18(250 mm×4.6 mm,5μm),流动相为乙腈-水(65∶35),流速为1.0 mL/min,柱温为35℃,检测波长为322 nm,进样量10μL。结果:蛇床子素在15.936~63.744μg/mL浓度范围内与峰面积积分值呈良好线性关系(r=0.9999),平均回收率100.6%(n=6)。三批样品含量测定结果显示,本品每毫升含蛇床子素0.291 mg。结论:本方法专属性强,重现性好,结果准确,操作简便,可作为止痒消炎水中蛇床子素的含量测定方法。Objective:To establish the high performance liquid chromatography(HPLC)method for the determination of osthole in relieving itching anti-inflammation fluid.Methods:HPLC was used with osthole as the reference substance.The chromatographic column was ZORBAX Eclipse Plus C18(250 mm×4.6 mm,5μm).The mobile phase was acetonitrile-water(65∶35),the flow rate was 1.0 mL/min,the column temperature was 35℃,and the detection wavelength was 322 nm,with injection volume of 10μL.Results:The linear range of osthol was from 15.936μg/mL to 63.744μg/mL(r=0.999,9),and the average recovery was 100.6%(n=6).The average content of osthol in three batches of samples was 0.291 mg/mL.Conclusion:This method is highly specific,reproducible,reliable and simple,which can be used as the method for the determination of osthole in relieving itching anti-inflammation fluid.
