摘  要:目的探讨白藜芦醇(Res)对神经元缺氧复氧(H/R)过程中细胞凋亡、氧化应激反应的影响。方法培养PC12细胞并诱导分化为神经元,将神经元分为对照组(无血清培养基+常氧处理)、H/R组(无血清培养基+缺氧复氧处理)和不同浓度Res组(80,40μmol·L^-1 Res+缺氧复氧处理),处理后测定细胞增殖活力、氧化应激产物含量及线粒体凋亡分子表达量。结果复氧后12,24,48 h,对照组细胞增殖活力明显高于H/R组、40μmol·L^-1 Res组及80μmol·L^-1 Res组,80μmol·L^-1 Res组高于40μmol·L^-1 Res组和H/R组,40μmol·L^-1 Res组高于H/R组,均差异有统计学意义(均P<0.05);复氧后48 h,对照组ROS、MDA、AOPP、8-OHdG含量及Bax、caspase-3、BaxmRNA、caspase-3mRNA表达量明显低于H/R组、40μmol·L^-1 Res组及80μmol·L^-1 Res组,80μmol·L^-1 Res组低于40μmol·L^-1 Res组和H/R组,40μmol·L^-1 Res组低于H/R组,均差异有统计学意义(均P<0.05);对照组Bcl-2、Bcl-2mRNA表达量均明显高于H/R组、40μmol·L^-1 Res组及80μmol·L^-1 Res组,80μmol·L^-1 Res组高于40μmol·L^-1 Res组和H/R组,40μmol·L^-1 Res组高于H/R组,均差异有统计学意义(均P<0.05)。结论Res能抑制神经元H/R过程中细胞凋亡和氧化应激反应。Objective To study the effects of resveratrol(Res)on apoptosis and oxidative stress in neurons during hypoxia-reoxygenation(H/R).Methods PC12 cells were cultured and induced to differentiate into neurons.They were divided into control group(no serum medium+normoxia treatment),H/R group(no serum medium+hypoxia/reoxygenation treatment),and different concentrations of Res group(40 or 80μmol·L^-1 resveratrol+hypoxia reoxygenation treatment),the cell proliferation activity,oxidative stress product content and mitochondrial apoptosis molecule expression were measured after treatment.Results At 12,24,and 48 h after reoxygenation,the cell proliferation activity of the control group was significantly higher than that of the H/R group,40μmol·L^-1 Res group,and 80μmol·L^-1 Res group,and the 80μmol·L^-1 Res group was higher than the 40μmol·L^-1 Res group and the H/R group,and the 40μmol·L^-1 Res group was higher than the H/R group,and the differences were statistically significant(P<0.05).The content of ROS,MDA,AOPP,8-OHdG and the expression of Bax,caspase-3,Bax mRNA,and caspase-3 mRNA in the control group were significantly lower than that in the H/R group,40μmol·L^-1 Res group,and 80μmol·L^-1 Res group.The 80μmol·L^-1 Res group was lower than the 40μmol·L^-1 Res group and the H/R group,and the 40μmol·L^-1 Res group was lower than the H/R group,the differences were statistically significant(P<0.05).The expression levels of Bcl-2 and Bcl-2 mRNA in the control group were significantly higher than those in the H/R group,40μmol·L^-1 Res group and 80μmol·L^-1 Res group.The 80μmol·L^-1 Res group was higher than the 40μmol·L^-1 Res group and the H/R group,and the 40μmol·L^-1 Res group was higher than the H/R group.The differences were statistically significant(P<0.05).Conclusion Resveratrol can inhibit the apoptosis and oxidative stress of neurons during hypoxia and reoxygenation.
