摘  要:目的观察表没食子儿茶素没食子酸酯(Epigallocatechin gallate,EGCG)对失血性休克大鼠炎症反应的抑制作用,探讨TLR4/Myd88信号通路的作用机制。方法SD大鼠30只,随机分为假手术组(Sham)、失血性休克(Hematogenic shock,HS)组、EGCG治疗组;HS和EGCG组大鼠均采用35%放血法建立HS模型,EGCG组于HS发生后腹腔注射EGCG(50mg/Kg)。HS发生4h后处死各组大鼠,HE染色观察肺组织病理学变化,ELISA检测炎性因子TNF-α、IL-6、IL-10浓度变化,Western blot及qPCR检测TLR4/Myd88信号通路相关蛋白表达情况。结果大鼠HS组,肺组织损伤严重,病理学可见肺间隔增宽,肺泡萎陷,大量炎性细胞浸润,肺功能损伤严重,动脉血气分析提示PaCO2明显升高,PaO2明显降低,血清炎性因子TNF-α、IL-6水平升高,IL-10水平降低;EGCG干预后,大鼠肺组织病理学形态和肺功能明显得到改善,TNF-α、IL-6,PaCO2水平明显降低,IL-10、PaO2水平明显升高,EGCG下调肺组织中TLR4、Myd88、NF-κB水平表达。结论EGCG明显改善HS大鼠肺功能及肺组织损伤,与调控TLR4/Myd88信号通路抑制大鼠炎性反应相关。Objective To investigate the inhibitory effect of epigallocatechin gallate(EGCG)on the inflammatory response in rats with hemorrhagic shock and to explore its relationship with TLR4/Myd88 signaling pathway.Methods Sprague-Dawley rats were randomly divided into sham operation group(Sham),hemorrhagic shock group(HS)and EGCG group.The pathological changes of lung tissue were observed by HE.The contents of TNF-α,IL-6 and IL-10 in serum were detected by ELISA.The expression of TLR4/Myd88 was detected by Western blot and qPCR.Results The severe lung injury,reduction of lung function and elevated inflammatory factors in HS group were found.EGCG lung decreased tissue damage and improved lung function,inhibited inflammatory factors,and down-regulated the expression levels of TLR4,Myd88 and NF-κB.Conclusion The effect of EGCG on the inflammatory response in rats with hemorrhagic shock is related to TLR4/Myd88 signaling pathway.
