摘 要:目的:观察蜜炼川贝枇杷膏体外抗甲型流感病毒的作用。方法:建立H1N1流感病毒感染鸡胚、MDCK细胞模型,以最大无毒浓度、细胞病变效应和噻唑蓝测定蜜炼川贝枇杷膏对鸡胚和细胞的毒性作用,以鸡红细胞凝集抑制试验观察药物的体外抗病毒作用。结果:与模型对照组比较,在鸡胚模型中,蜜炼川贝枇杷膏86、172 mg/ml预防用药可一定程度上抑制病毒复制(抑制率分别为16.7%、50%),治疗用药22、43、86 mg/ml能明显抑制病毒复制,86 mg/mL时抑制率为100%;在MDCK细胞模型中,蜜炼川贝枇杷膏在预防、直接杀伤和治疗用药时均有显著抑制病毒的作用,对病毒的100%抑制剂量分别为40 mg/mL、40 mg/mL和20 mg/mL;治疗作用优于预防作用。结论:蜜炼川贝枇杷膏对H1N1流感病毒有一定的预防作用和明显的治疗作用。Objective: To investigate the antiviral activity of Milian Chuanbei Pipa Gao against influenza virus(H1 N1) infection in vitro. Methods:The chicken embryo and MDCK cells models infected by influenza A H1 N1 virus were established. Its toxicity was determined by maximum non-cytotoxic concentration, cytopathic effect and MTT assay. The antivirus effect on H1 N1 in vitro was observed by hemagglutination inhibition test. Results: Compared with the model group, for chicken embryo model,the virus replications(inhibition rate16.7%、50%) were inhibited in Milian Chuanbei Pipa Gao prophylactic groups(86、172 mg/ml), in therapeutic groups(22、43、86 mg/ml) the virus replications also were significantly inhibited(inhibition rate 100% in 86 mg/mL group). For MDCK cells model, virus replication was significantly inhibited in Milian Chuanbei Pipa Gao prophylactic group(100% inhibition rate at a dose of 40 mg/mL), in therapeutic groups(100% inhibition rate at a dose of 20 mg/mL), it showed the direct killing effect(100% inhibition rate at a dose of 40 mg/mL). While its therapeutic effect was more significant than the preventive effect. Conclusion: Milian Chuanbei Pipa Gao shows certain preventive effect and obvious therapeutic effect on influenza virus(H1 N1) in vitro.
