摘 要:目的:探究商陆皂苷对脂多糖诱导的小鼠乳腺炎的效果及作用机制。方法:将BALB/c雌性小鼠随机分成正常组、模型组、地塞米松组(DEX,5.0 mg·kg^-1)和商陆皂苷给药组(50,25,12.5 mg·kg^-1),采用脂多糖(LPS)诱导产后雌小鼠(BALB/c)乳腺炎模型,分析小鼠乳腺组织的病理情况和中性粒细胞髓过氧化物酶(MPO)活性水平;测定乳腺组织中炎症因子的含量及氧化应激水平;通过蛋白免疫印迹法(Western blot)评估商陆皂苷对Toll样受体4(TLR4)/核转录因子-κB(NF-κB)信号通路蛋白的影响。结果:与正常组比较,模型组的乳腺组织中有充血和瘀血等典型的乳腺炎变化,MPO水平升高,肿瘤坏死因子-α(TNF-α),白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)的表达水平显著升高(P<0.01);与LPS模型组比较,各商陆皂苷组能够显著改善小鼠乳腺组织的炎性损伤,降低嗜中性粒细胞的分泌,降低MPO的活性,促炎症因子TNF-α,IL-1β和IL-6的表达水平也明显下调,;升高超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)酶活力,同时降低丙二醛(MDA)的水平(P<0.05,P<0.01),降低TLR4/NF-κB信号通路蛋白的表达(P<0.05,P<0.01)。结论:商陆皂苷对脂多糖诱导的小鼠乳腺炎损伤有保护作用,其可能与抑制TLR4/NF-κB信号通路蛋白的表达有关。Objective:To explore the effect and mechanism of esculentoside on lipopolysaccharideinduced mastitis in mice.Method:Female BALB/c mice were randomly divided into control group,model group,dexamethasone group(DEX,5.0 mg·kg^-1)and esculentoside group(50,25,12.5 mg·kg^-1).The mastitis model of postpartum female mice(BALB/c)induced by lipopolysaccharide(LPS)was used to analyze the pathological conditions of breast tissue and the activity of recombinant myeloperoxidase(MPO),factor content and oxidative stress level.Western blot was used to evaluate the effect of esculentoside on Toll-like receptor4(TLR4)/nuclear transcription factor(NF-κB)signaling pathway proteins.Result:Compared with the normal group,the breast tissue of the model group had typical mastitis changes,such as hyperemia and congestion,the level of MPO increased,and the expression levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)increased significantly(P<0.01).Compared with LPS model group,esculentoside groups could significantly improve the inflammatory damage of mammary gland tissue,reduce the secretion of neutrophils and the activity of MPO,the expression levels of pro-inflammatory factors TNF-α,IL-1βand IL-6 were also significantly down-regulated,the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were increased,while the level of malondialdehyde(MDA)was decreased,and the activation of TLR4/NF-κB signaling pathway was inhibited by esculentoside(P<0.05,P<0.01).Conclusion:Esculentoside have a protective effect on lipopolysaccharide-induced mastitis in mice,which may be related to the inhibition of TLR4/NF-κB signaling pathway protein expression.
