摘  要:目的考察雷公藤"以药制药"炮制减毒作用机制。方法雷公藤生品及其"以药制药"炮制品被给予生理小鼠14 d,通过对血清、肝、肾生物样本的分析,评价其炮制减毒作用机制。结果雷公藤生品引起了小鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、肌酐(Cr)和尿素氮(BUN)、肝和肾的脂质过氧化产物丙二醛(MDA)以及炎性介质肿瘤坏死因子(TNF)-α水平的极显著升高,肝和肾的抗氧化物谷胱甘肽(GSH)、谷胱甘肽-S-转移酶(GST)、谷胱甘肽过氧化物酶(GPx)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)以及抗炎性介质白介素(IL)-10水平的显著降低;与雷公藤生品组比较,金银花制雷公藤(JYHLGT)、白芍制雷公藤(BSLGT)、金钱草制雷公藤(JQCLGT)、甘草制雷公藤(GCLGT)、绿豆制雷公藤(LDLGT)等5个炮制品组均不同程度地逆转了以上指标的异常。结论金钱草、金银花、绿豆、白芍、甘草"以药制药"炮制雷公藤均可起到减毒作用,其减毒作用机制可能与抑制脂质过氧化、增强抗氧化以及抑制炎症反应有关。Objective To investigate herbs-processed detoxication mechanisms of Radix Tripterygium wilfordii(LGT)in physiological mice.Method LGT raw and herbs-processed products were given to physiological mice for 14 days,and the serum,liver and kidney were analyzed to evaluate the detoxication effects and mechanisms.Results LGT raw product administration caused significant elevation in serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),creatinine(Cr)and urea nitrogen(BUN),levels of lipid peroxidation product malondialdehyde(MDA)and inflammatory mediator tumor necrosis factor(TNF)-αin liver and kidney as well as significant reduction in glutathione(GSH),glutathione-s transferase(GST),glutathione peroxidase(GPx),superoxide dismutase(SOD),catalas(CAT)and interleukin(IL)-10 in liver and kidney of mice.Compared with the LGT raw product group,administration of the five processed products including Jinyinhua(Lonicerae Japonicae Flos)-processed LGT(JYHLGT),Baishao(Paeoniae Radix Alba)-processed LGT(BSLGT),Jinqiancao(Lysimachiae Herba)-processed LGT(JQCLGT),Gancao(Glycyrrhizae Radix et Rhizoma)-processed LGT(GCLGT),and mung bean-processed LGT(LDLGT)reversed the anomalies of the above indicators to varying degrees.Conclusion Processing with medicinal herbs(JQC,JYH,LD,BS and GC)exerted detoxication effects on LGT-induced hepatotoxicity and nephrotoxicity,and the mechanisms could be related to inhibition of lipid peroxidation,enhancement of antioxidant activity,and inhibition of inflammatory response.
