摘  要:目的通过给予慢传输型便秘大鼠增液汤干预治疗,研究其对慢传输型便秘(STC)大鼠结肠中AQP3含量的影响及血清中NOS水平的影响,探讨慢传输型便秘的发病机制及增液汤润肠通便的作用机制。方法将60只SD大鼠按随机数字表法分为6组,即:正常组,模型组,伊托必利组,增液汤低剂量组,增液汤中剂量组,增液汤高剂量组,除正常组外,其余各组均给予复方地芬诺酯建立慢传输型便秘大鼠模型,造模成功后,分别给予相应药物灌胃治疗,观察增液汤对慢传输型大鼠的通便作用。采用免疫组织化学法检测各组AQP3在结肠的分布和表达情况,测定其平均光密度(MOD)值,并取最终均值代表AQP3阳性表达参数;采用Q-PCR法检测AQP3在结肠组织中的水平,蛋白免疫印迹法(Western Blot)检测AQP3蛋白表达情况。用ELISA检测大鼠血清中NOS的水平变化。结果与正常组相比,模型组近端结肠组织中AQP3含量明显增加(P<0.05),经增液汤及伊托必利治疗后,与模型组相比,增液汤中,高剂量组及伊托必利组的近端结肠AQP3含量明显下降(P<0.05),差异有统计学意义,增液汤低剂量近端结肠AQP3含量有下降趋势但差异无统计学意义。ELISA检测血清中NOS的浓度,模型组大鼠血清中NOS浓度较正常组相比明显升高(P<0.05),差异有统计学意义;经增液汤及伊托必利治疗后,与模型组相比,增液汤低、中、高剂量组及伊托必利组的血清中NOS含量均明显下降(P<0.05),差异有统计学意义,其中伊托必利组与增液汤低剂量组比较(P>0.05),差异无统计学意义。结论近端结肠组织中AQP3含量增加可能是STC的发病机制之一;增液汤通过下调AQP3表达可能是其发挥"增液行舟"效应从而治疗STC的作用机制之一。Objective To explore the pathogenesis of slow transit constipation by studying the effect of Zengye Decoction on AQP3 in slow transit constipation(STC)ratsandprovide the evidence for studying themechanism of traditional Chinese medicine.Methods Sixty SD rats were randomly divided into six groups:normal group,model group,Zengye Decoction high,middle and low dose groups,itopride group.Except the normal group,the other groups were given diphenoxylate by gavage to induce the constipationmodel.Aftermodeling,the rats were given the corresponding drugs by gavage.We observed thelaxative effect of Zengye Decoction on slow transit constipation rats.The immunohistochemical method was used to detect the AQP3 expression in the colon and the average optical density(MOD)value,and ultimately chosethe average value as the positive expression of AQP3 parameter.Q-PCR method was used to detect AQP3 levelin colonic tissue.Western blotting was used to detect the expression of AQP3 protein.The level of NOS in the serum of rats was detected by ELISA.Results Compared with the normal group,the model group’scontent of AQP3 in the proximal end of the colon tissue was increased significantly(P<0.05).After Zengye Decoction and itopride treatment,compared with the model group,Zengye Decoctionhigh dose group and itopride group’s AQP3 content in the proximal end of colonwas significantly decreased(P<0.05)and there was a significant difference.The Zengye Decoctionlow dose group’s AQP3 content in the proximal end of colon was decreased but the difference was not statistically significant.The concentration of NOSin serum was tested by ELISA.The concentration of NOS in the serum of rats in the model group increased significantly compared with that of the normal group and the difference was statistically significant(P<0.05).After Zengye Decoction and itopridetreatment,compared with the model group,Zengye Decoctionlow,middle and high dose groups and itopride group’sNOS contents were decreased significantly and the difference was statistically
