溪黄草下调miR-1290表达抑制结肠癌细胞恶性行为的研究

摘　　要：目的研究中药溪黄草对结肠癌细胞中miR-1290表达、以及结肠癌细胞恶性行为的影响。方法制备溪黄草乙醇提取物,以不同浓度(20~80μg/mL)作用于体外培养的人结肠癌细胞HCT116,采用实时荧光定量PCR法检测细胞内和细胞培养液中miR-1290的表达水平,采用CCK-8法检测细胞增殖能力,采用划痕法检测细胞迁移能力,采用Matrigel穿膜法检测细胞侵袭能力。结果溪黄草乙醇提取物显著下调了HCT116细胞内和细胞培养液中miR-1290的表达水平,有力抑制了HCT116细胞的增殖能力、迁移能力和侵袭能力。结论中药溪黄草能够下调结肠癌细胞对促癌性小RNA miR-1290的表达和分泌,抑制结肠癌细胞的恶性行为,在结肠癌的治疗上具有良好的应用价值。Objective To investigate the influence of Serrate rabdosia on the miR-1290 expression and the malignant behaviors of colon cancer cells.Methods HCT116 human colon cancer cells were cultured and treated with different concentrations(20~80μg/mL)of Serrate rabdosia ethanol extract.The expression of miR-1290 in HCT116 cell and that in the cell culture medium were detected by Realtime PCR.The cell proliferation was measured by CCK-8 assay,the cell migration determined by scratch wound assay,and cell invasion evaluated by Matrigel assay.Results The Serrate rabdosia alcohol extract significantly down-regulated miR-1290 expression in HCT116 cells and that in the cell culture medium.It strongly inhibited the proliferation,migration and invasion of HCT116 cells.Conclusion The Chinese herb Serrate rabdosia inhibits the malignant behaviors of colon cancer cells by down-regulating the expression and secretion of the oncogenic non-coding small RNAmiR-1290,and thus is valuable for the treatment of colon cancer.
