摘  要:磷脂酰肌醇-4-磷酸3-激酶催化亚基2型α(phosphatidylinositol-4-phosphate 3-kinase,catalytic subunit type 2 alpha,PIK3C2A)基因在生物体中主要参与细胞代谢、细胞增殖以及细胞内蛋白质的转移,并与蛋白质合成以及肌肉质量有关。本研究分析宁海黄鸡和广西黄鸡2个国内优质地方鸡种PIK3C2A基因多态性及其与屠体性状之间的相关性,以期为标记辅助选择提供理论依据。应用鸡高密度(600 K)SNP芯片对PIK3C2A基因中的单核苷酸多态性(SNP)进行分析,在总共被检测出的42个SNPs中,有4个SNPs(rs312835349、rs13757033、rs16466818和rs317679804)位于PIK3C2A基因的外显子区域,且这4个SNPs共显示出3种基因型(AA、AB和BB),并呈现出中度多态性;在PIK3C2A基因内含子区域的38个SNPs中,发现宁海黄鸡的7个SNPs和广西黄鸡的6个SNPs分别与部分屠体性状显著相关(P<0.05或P<0.01),其中有3个SNPs为2个鸡品种共有(C939A、T6477C和A38680G),推测这3个SNPs可能是鸡标记辅助选种的潜在候选遗传标记。研究结果表明,PIK3C2A基因可能是影响鸡屠体性状的关键基因之一,可作为分子标记辅助选择的候选基因。PIK3C2A(phosphatidylinositol-4-phosphate 3-kinase,catalytic subunit type 2 alpha)gene is mainly involved in cell metabolism,cell proliferation,and intracellular protein transfer in organisms,and is related to protein synthesis and muscle quality.The object of this study was to assess the association of polymorphisms in PIK3C2A gene with carcass traits in two Chinese chicken breeds(Ninghai and Guangxi yellow chickens).Initially,Single nucleotide polymorphisms(SNPs)in PIK3C2A gene were identified and analyzed based on the 600 K chicken genotyping array.Of the total 42 identified SNPs,only 4 SNPs were found in exon regions of PIK3C2A gene.These SNPs revealed 3 genotypes(AA,AB and BB)and presented moderate polymorphisms in these two chicken breeds.Seven SNPs of PIK3C2A gene in Ninghai yellow chickens and 6 SNPs in Guangxi yellow chickens were significantly associated with carcass traits(P<0.05 or P<0.01),whereas all of these described SNPs are located within intron.The two chicken populations possessed 3 common SNPs(C939 A,T6477 C and A38680 G)that significantly associated with carcass traits,indicating that these 3 SNPs could be a potential candidate genetic marker for marker-aid breeding in chickens.Our results suggest that the PIK3C2A gene may be one of the causative genes that control carcass traits in chicken breeds,which could be the candidate genes of molecular marker-assisted selection.
