摘 要:目的研究脂肪干细胞移植对绝经后骨质疏松模型大鼠骨密度及骨组织形态学影响。方法将3个月龄雌性SD大鼠随机分为假手术组(110只)、骨质疏松组(110只)、脂肪干细胞移植组(116只)。假手术组摘除卵巢周围的脂肪组织;骨质疏松组、脂肪干细胞移植组摘除双侧卵巢组织;脂肪干细胞移植组给予2×107个脂肪干细胞尾静脉注射、假手术组和骨质疏松组给予等量生理盐水尾静脉注射,每周1次,8周后检测大鼠的骨密度、骨组织形态变化。结果与假手术组比较,骨质疏松组大鼠股骨的骨密度明显降低(P<0.05);与骨质疏松组比较,脂肪干细胞移植组大鼠股骨的骨密度明显升高(P<0.05)。与假手术组比较,骨质疏松组大鼠右侧股骨的骨小梁面积百分比、骨小梁厚度明显降低,骨小梁间距明显增加(P<0.05);与骨质疏松组比较,脂肪干细胞移植组大鼠右侧股骨的骨小梁面积百分比、骨小梁厚度明显增加,骨小梁间距明显降低(P<0.05);与假手术组比较,骨质疏松组大鼠左侧股骨中矿物质盐的含量明显降低(P<0.05);与骨质疏松组比较,脂肪干细胞移植组大鼠左侧股骨中矿物质盐的含量明显增加(P<0.05)。结论脂肪干细胞移植能够改善绝经后骨质疏松大鼠的骨密度及骨形态学指标。Objective To study the effects of adipose-derived stem cells transplantation on bone mineral density and bone histomorphology in postmenopausal osteoporosis rats.Methods Three-month-old female Sprague-Dawley(SD)rats were randomly divided into sham operation group,osteoporosis group and adipose-derived stem cell group,with 10 rats in each group.The adipose tissue around the ovary was removed in the sham operation group,and the bilateral ovarian tissue was removed in the osteoporosis group and adipose-derived stem cells.2×107 adipose-derived stem cells were injected into the caudal vein in the adipose-derived stem cells group and the same amount of normal saline was injected into the caudal vein in the sham operation group and the osteoporosis group once a week.After 8 weeks,the changes of bone mineral density and bone histomorphology of the rats were measured.Results Compared with the sham operation group,the bone mineral density of femur in osteoporosis group was decreased significantly(P<0.05).Compared with the osteoporosis group,the bone mineral density of femur in adipose stem cell group was increased significantly(P<0.05).Compared with the sham operation group,the trabecular area percentage and trabecular thickness of the right femur in the osteoporosis group were decreased significantly,and the trabecular spacing was increased significantly(P<0.05).Compared with the osteoporotic group,the trabecular area percentage and trabecular thickness of the right femur in the adipose stem cell group were increased significantly,and the trabecular spacing was decreased significantly(P<0.05).Compared with the sham operation group,the content of mineral salt in the left femur of the osteoporosis group was significantly lower(P<0.05),and that of adipose-derived stem cellsl group was significantly higher than that of the osteoporosis group(P<0.05).Conclusion Adipose-derived stem cells transplantation can improve the bone mineral density and thebone morphology in postmenopausal osteoporotic rats.
