摘 要:酶软化过程中,常常使用非离子表面活性剂促进酶的渗透。本文采用紫外、荧光、园二色谱等光谱学方法,考察了胰蛋白酶在非离子表面活性剂平平加O的存在下活性变化及结构稳定性。研究发现平平加O的存在下胰蛋白酶的活性保持稳定并有一定的激活作用,紫外和荧光光谱研究发现平平加O的存在不会改变胰酶蛋白的三级结构。圆二色谱分析发现,随着平平加浓度的增加,胰酶蛋白的二级结构变化较小。本研究显示在胰酶软化过程中添加非离子表面活性剂平平加O,对胰酶的活性和结构均能维持稳定。In Bating process, nonionic surfactants commonly applied for help the penetration of trypsin. In this paper, the structure stability and activity of trypsin interacted by peregal O were investigated by spectroscopic methods, including UV spectroscopy, fluorescence spectroscopy,and circular dichroism spectroscopy. The results showed that the structure stability and activity of trypsin was maintained with the addition of peregal O. The activity of trypsin was slightly activated by the addition of peregal O, and the secondary and the tertiary structures of trypsin was preserved. It illustrated that the addition of peregal O in the bating process will be no harm to trypsin.
