摘　　要：制备合格的培养基是顺利开展植物组织培养试验的基础,在本科毕业实习生开展毕业论文试验过程中,制备的固体培养基软硬不一、混合不匀是最为突出的问题之一。对制备好的培养基表现偏软或过硬以及软硬不匀的判断标准、产生的原因及对应的解决措施进行了重点阐述。此外,对培养基的成分组成、pH值、灭菌效果等在不同处理组间或处理组内的不匀表现产生的原因进行了分析,并提出相应的解决方法,以期为学生顺利开展植物组培试验,制备合格的固体培养基提供参考。The preparation of qualified media was the basis for plant tissue culture.The problem of inhomogeneity of medium,such as incomplete mixing,compositional fluctuation was the most prominent problem in the preparation of solid medium in the course of graduation thesis research.In this article,the criteria for judgment,the causes and the corresponding solutions of too soft or hard performances of prepared solid medium were expounded.Besides,the performances of inhomogeneous characteristics of medium,such as the medium composition,the sterilizing effect and pH value of solid medium existed in medium preparation were presented.The causes for these problems were analyzed,and the improvement methods or solutions were put forward.The paper would provide reference for preparing qualified solid media in plant tissue culture research for the undergraduates.
