摘  要:目的以黄酮为检测指标进行银杏愈伤组织培养条件筛选研究。方法采用MS培养基,对银杏叶茎等进行初代愈伤组织的诱导,按照中国药典(2015版)规定的方法进行检测黄酮含量,通过不同配方的培养基的对比试验筛选出最佳配方,在确定最佳配方之后再加以继代培养。结果其中初代培养最佳配方为MS+3.0 mg/L6-BA+2.0 mg/LNAA+3%蔗糖+0.7%琼脂。愈伤生长速度呈S型增长,但在45 d左右顶部易褐化。在不同外植体的愈伤能力比较中,茎段的愈伤能力最强,叶柄次之,叶片最弱,但叶片后期生长状况最好。结论3.0 mg/L的6-BA与2.0 mg/LNAA的培养基最有利于黄酮的合成的最佳条件。Objective To study the callus culture of Ginkgo biloba and the extraction and detection of its flavonoids.Methods MS medium was used to induce primary callus of Ginkgo biloba leaves,stems and so on.The best formula was screened out through the contrast test of different media.After determining the best formula,subculture was added.Results The best formula for primary culture was MS+3.0mg/L6-BA+2.0mg/LNAA+3%sucrose+0.7%agar.The growth rate of callus was S-shaped,but the top of callus was browning at about 45 days.In comparison of callus ability of different explants,stem segment had the strongest callus ability,petiole was the second,and leaf was the weakest,but leaf growth condition was the best in later stage.The extraction and detection of Flavonoids from callus were carried out in strict accordance with Chinese Pharmacopoeia(2015 Edition).Conclusion The results showed that the culture medium of 3.0mg/l 6-BA and 2.0mg/lnaa was the most favorable for the synthesis of flavonoids.
