摘 要:目的探讨血管紧张素1-7(angiotensin1-7,Ang1-7)对肥厚心肌总缝隙隙连接蛋白43(total connexin43,Total-CX43)和CX43丝氨酸残基368磷酸化位点(phosphorylation of serine residue 368-CX43,PS368-CX43)表达的影响。方法选择5只16周龄雄性WKY(normotensive wistar kyoto,WKY)大鼠作为正常血压组,10只16周龄自发性高血压大鼠(spontaneous hypertension rat,SHR)作为SHR组。然后将10只SHR随机分为安慰剂组(SHR+NS)和Ang1-7干预组(SHR+A),3组在干预前后分别测定尾动脉收缩压(systolic blood pressure,SBP)明确血压变化,干预28 d后行HE染色比较各组之间心肌肥厚程度,免疫组化及Western blot检测Total-CX43和PS368-CX43的表达。结果与WKY组相比,干预前SHR+NS组和SHR+A组血压均升高(P<0.01)。干预后与WKY组相比,SHR+NS组和SHR+A组大鼠血压均升高(P<0.01),与SHR+NS组相比,SHR+A组大鼠血压下降(P<0.01)。HE染色结果显示WKY组大鼠心肌排列整齐,组织结构清晰;SHR+NS组大鼠心肌排列紊乱,组织结构欠清晰;SHR+A组介于二者之间。免疫组化检测Total-CX43和PS368-CX43分布,与WKY组相比,SHR+NS和SHR+A组均降低(P<0.05);与SHR+NS组相比,SHR+A组增加(P<0.01)。Western blot结果显示,与WKY组相比,SHR+A组大鼠Total-CX43和PS368-CX43降低(P<0.01)。与SHR+NS组相比,SHR+A组大鼠Total-CX43和PS368-CX43升高(P<0.01)。结论在高血压大鼠模型中,Ang1-7可降压、影响肥厚心肌Total-CX43/PS368-CX43的表达。Objective To investigate the effects of angiotensin 1-7(Ang1-7)on the expression of Total connexin43(Total-CX43)and phosphorylation of serine residue 368-CX43(PS368-CX43)in hypertrophic myocardium.Methods Five 16-week-old male normotensive wistar kyoto rats(WKY)were selected as the normal blood pressure group,and ten 16-week-old spontaneous hypertension rats(SHR)were selected as the SHR group.Then,10 SHRS were randomly divided into the placebo group(SHR+NS)and the Ang1-7 intervention group(SHR+A).Systolic blood pressure(SBP)of the ciliary artery was measured before and after the intervention to determine the changes in blood pressure.After 28 days of intervention,HE staining was performed to compare the degree of myocardial hypertrophy between the groups,and the expression of Total-CX43 and PS368-CX43 was detected by immunohistochemistry and Western blot.Results Compared with WKY group,blood pressure in SHR+NS group and SHR+A group before intervention was significantly increased(P<0.01).After intervention,compared with WKY,the blood pressure of SHR+NS group and SHR+A group increased significantly(P<0.01),but compared with SHR+NS group,the blood pressure of SHR+A group decreased significantly(P<0.01).The results of HE staining showed that the myocardium of WKY group was arranged neatly with clear tissue structure,while that of SHR+NS group was disordered with unclear tissue structure,and that of SHR+A group was between the two.Immunohistochemical analysis showed the distribution of Total-CX43 and PS368-CX43.Compared with WKY group,SHR+NS and SHR+A were significantly decreased(P<0.05),while compared with SHR+NS group,SHR+A was significantly increased(P<0.01).Western blot results showed that Total-CX43 and PS368-CX43 were decreased in SHR+A group compared with WKY group(P<0.01).Compared with SHR+NS group,Total-CX43 and PS368-CX43 in SHR+A group increased(P<0.01).Conclusion In the hypertensive rat model,Ang1-7 could reduce blood pressure and affect the expression of Total-CX43/PS368-CX43 in the hypertrophic myocardiu
