摘  要:目的研究脂肪乳对布比卡因致原代海马神经元细胞毒性作用的影响。方法选用原代培养第9天的SD大鼠海马神经元细胞,将细胞分为4组:空白对照组(Control组)不进行任何处理;布比卡因组(BPV组)加入1 mmol·L^-1布比卡因;脂肪乳剂组(LE组)加入1%脂肪乳剂;布比卡因+脂肪乳剂组(BPV+LE组)加入1 mmol·L^-1布比卡因和1%脂肪乳剂。4组细胞均处理24 h后分别采用CCK-8法检测细胞活力和电生理膜片钳技术对各组细胞的动作电位进行检测分析。采用全细胞记录法分别对动作电位幅度、动作电位时程和动作电位幅度对应的峰值进行对比分析。结果与Control组比较,BPV组海马神经元细胞活力减弱(P<0.05)。与BPV组比较,LE组和BPV+LE组海马神经元细胞活力增强(P<0.05)。4组间细胞的动作电位幅度和动作电位幅度对应的峰值差异无统计学意义(P>0.05)。与Control组比较,BPV组细胞的动作电位时程增长(P<0.05)。与BPV组比较,BPV+LE组细胞的动作电位时程缩短(P<0.05)。结论脂肪乳剂可通过增加细胞活力和影响细胞动作电位的时程来减轻布比卡因引起的海马神经细胞毒性。Objective To study the effect of lipid emulsion on the cytotoxicity of primary cultured hippocampus neurons induced by bupivacaine.Methods The primary cultured hippocampal neuron cells of SD rats of the 9th day were divided into four groups:blank control group(Control group)without any treatment;Bupivacaine group(BPV group)with 1mmol·L^-1 bupivacaine;fat emulsion group(LE group)with 1%fat emulsion;bupivacaine+fat emulsion group(BPV+LE group)with 1mmol·L^-1 bupivacaine and 1%fat emulsion.CCK-8 method and electrophysiological patch clamp technique were used to detect the action potential of cells in each group.The peak values of action potential amplitude,action potential duration and action potential amplitude were compared and analyzed by whole cell recording method.Results Compared with the Control group,the activity of hippocampal neurons in BPV group decreased significantly and the difference was statistically significant(P<0.05).Compared with the BPV group,the hippocampal neuron cells in the LE group and the BPV+LE group were significantly more active and statistically significant(P<0.05).There was no significant difference between the action potential amplitude and the peak amplitude of the four groups of cells(P>0.05).Compared with the Control group,the action potential duration of cells in the BPV group increased,and the difference was statistically significant(P<0.05).Compared with the BPV group,the action potential duration of cells in the BPV+LE group was shortened,and the difference was statistically significant(P<0.05).Conclusion Lipid emulsion can reduce the toxicity of bupivacaine to hippocampal neurons by increasing cell viability and affecting the time course of cell action potential.
