摘 要:GATA转录因子在植物中广泛存在,并参与生长发育、生物和非生物胁迫等多种生物学过程,为深入阐明GATA基因家族在剑麻相关生物学过程中的重要功能。本研究基于转录组水平鉴定手段,对剑麻基因家族进行鉴定分析以及表达模式研究。在前期已完成的RNA-seq数据上,利用相关软件对剑麻GATA基因家族进行鉴定,同时对得到的剑麻GATA基因进行理化性质、保守结构域、进化关系和表达模式分析。在剑麻中共筛选获得23个GATA基因,其编码蛋白质的氨基酸数目在118~581之间,分子量约为12.74~64.87 ku,等电点为5.19~10.22。亚细胞定位预测结果显示18个AhGATAs定位于细胞核,3个AhGATAs定位于叶绿体,2个定位于线粒体。AhGATAs基因家族含有GATA、RPN13_C、AhXH、CCT和Tify这5个相对保守的结构域。进化树分析表明剑麻GATA蛋白可分为5个亚家族。23个剑麻GATA基因在剑麻紫色卷叶病不同发病时期呈现不同的表达规律。本研究在转录水平上初步完成了剑麻GATA基因家族的鉴定与功能分析,为后续深入解析AhGATAs基因在剑麻中的具体功能提供参考。GATA transcription factors exist widely in plants and participate in many biological processes,such as growth and development,biological and abiotic stresses.In order to further elucidate the important function of GATA gene family in the biological process of sisal.Based on the transcriptome level identification method,the GATA gene family of sisal was identified and analyzed,and the expression pattern was studied.Based on the previous completed RNA-seq data,the agave GATA gene family were identified by related softwares.The physicochemical properties,conserved domains,evolutionary relationships and expression patterns of the identified agave GATA gene were also analyzed.The results showed that 23 GATA genes were screened in agave,and the encoded amino acids number of AhGATAs genes were between 118 and 581,the molecular weight ranged from 12.74 to64.87 ku,and the isoelectric point varied from 5.19 to 10.22.Subcellular localization prediction showed that 18 AhGATAs were localized in the nucleus,3 AhGATAs were localized in the chloroplasts,and 2 AhGATAs were localized to mitochondria.The AhGATAs gene family contained five relatively conserved domains which were GATA,RPN13_C,AhXH,CCT and Tify.Phylogenetic tree analysis indicated that the agave GATA protein could be divided into five subfamilies.The 23 agave GATA genes showed different expression patterns in different stages of agave purple leaf curl disease.This study preliminarily completed the identification and functional analysis of the sisal GATA gene family in transcriptional level,laying a important foundation for further analysis of the specific functions of AhGATAs genes in agave.
