摘 要:热休克蛋白70(HSP70)在细胞保护等方面起着关键作用,但至目前为止,在日本三角涡虫(Dugesia japonica)中,HSP70的具体功能尚不明确.以再生能力极强的日本三角涡虫为实验材料,从转录组数据库中筛选了在再生过程中显著调高的Djhsp 70-4基因进行研究.实时荧光定量PCR结果显示:在受到外界刺激后,Djhsp 70-4基因的表达量显著上调.通过整体原位杂交技术,观察到Djhsp 70-4基因在整体涡虫身体两侧及再生芽基处有较强的杂交信号.利用RNA干扰技术敲低Djhsp 70-4基因的表达量后,整体及再生组涡虫出现头部溶解现象.综上所述,Djhsp 70-4基因可能在日本三角涡虫的稳态维持以及再生方面发挥重要作用.Heat shock proteins 70(HSP70)play a key role in cell protection.But so far,in Dugesia japonica,the specific functions of HSP70 are unclear.In this paper,D.japonica with extremely high regeneration ability is used as experimental material.From the transcriptome database,the Djhsp 70-4 gene significantly increased during the regeneration process were selected for research.The results of qRT-PCR showed that after being stimulated by the outside world,the expression of Djhsp 70-4 gene was significantly increased.Through the whole-in-situ hybridization technique,it was observed that the Djhsp 70-4 gene had a strong hybridization signal on both sides of the body of the whole planarian and at the regenerated blastma.After the RNA interference technique was used to reduce the expression of the Djhsp 70-4 gene,head dissolution occurred in the whole and regenerated planarians.In summary,the Djhsp 70-4 gene may play an important role in the maintenance and regeneration of D.japonica.
