摘 要:山西灵空山国家级自然保护区位于太岳山脉中段,近二百年来没有褐马鸡分布的记载和报道,2005年首次发现褐马鸡在此栖息。灵空山褐马鸡地理种群的来源和遗传背景的研究尚属空白。文章采用线粒体DNA分子标记技术,对灵空山褐马鸡种群的遗传多样性、遗传结构、系统发育进行了分析。结果显示:4只个体均获得长度为1238 bp的mtDNA D-loop全序列以及1458 bp Cyt b的全序列,分别定义了3个和2个单倍型。两条序列D-loop和Cyt b单倍型多样性(h)平均值分别为0.833和0.5,核苷酸多样性平均值(π)分别为0.00283和0.00206,平均遗传距离分别为0.003和0.002。反映了灵空山自然保护区褐马鸡种群具有较高的单倍型多样性和较低的核苷酸多样性,种群遗传多样性较低。这与该地区褐马鸡的资源现状、分布范围相适应。Brown-eared Pheasant(Crossoptilon mantchuricum)is a Chinese endemic endangered bird,which is listed as the first-grade State Protection Animal.Lingkong Mountain Nature Reserve is located in the middle of Taiyue Mountain,Shanxi,where no existence of Brown-eared Pheasant was reported in the past two hundred years until 2005.The origin and genetic background of the newly found Brown-eared Pheasant population need to be investigated.To study the genetic structure,polymorphism and maternal origin of the Brown-eared Pheasant population in Lingkongshan Nature Reserve,we obtained four Brown-eared Pheasant individuals and sequenced the mitochondrial DNA(mtDNA)D-loop and Cytochrome b(Cyt b)gene.We performed the analysis of genetic polymorphism,structure and phylogeny with the sequencing data.Using the 1238 bp D-loop and 1458 bp Cyt b sequences from all the four individuals,we defined 3 and 2 haplotypes,respectively.The average haplotype diversity(h)were 0.833 and 0.5,respectively.The average nucleotide diversity(π)were 0.00283 and 0.00206,respectively.The average genetic distance were 0.003 and 0.002,respectively.These results showed relatively low level of genetic diversity,which is consistent with the known abundance and distribution range of this species.
