摘  要:[目的]观察黄蒲通窍胶囊对VaD大鼠海马CA1区突触超微结构的影响。[方法]采用永久性结扎双侧颈总动脉的方法制作血管性痴呆大鼠模型。各组大鼠术后24 h给予相应灌胃:正常对照组、假手术组及模型组给予生理盐水灌胃,剂量为5.0 ml/(kg·d);多奈哌齐组给予多奈哌齐配制液灌胃,剂量为0.63 mg(/kg·d),连续45 d;黄蒲通窍胶囊组给予黄蒲通窍配制液灌胃,剂量为1.51 g(/kg·d),连续灌胃45 d。末次用药24 h后,应用免疫组织化学法观察各组大鼠海马CA1区染色情况,以电镜观察海马CA1区突触超微结构。[结果]模型大鼠海马CA1区突触超微结构:突触间隙各组无明显差异(P>0.05);与模型组比较,黄蒲通窍组突触后膜致密物(PSD)厚度、突触界面曲率和活性带长度均显著增加(P<0.01),与多奈哌齐组比较差异无统计学意义(P>0.05)。[结论]黄蒲通窍胶囊能够改善海马CA1区突触结构可塑性。[Objective]To observe the effect of Huangpu Tongqiao capsule on synaptic ultrastructure of hippo⁃campal CA1 region in VaD rats.[Methods]Rat models of vascular dementia were made by permanently ligating the bilateral common carotid arteries,and rats in each group were given intragastric administration 24 hours after operation:normal control group,sham operation group,and model group were given intragastric saline at a dose of 5.0 ml(/kg·d);Donepezil group was given Donepezil preparation solution 0.63 mg(/kg·d)for 45 days consecutive⁃ly;Huangpu Tongqiao capsule group was given Huangpu Tongqiao preparation solution 1.51 g(/kg·d)for 45 days consecutively;At 24 hours after the last administration,immunohistochemistry was used to observe the staining of hippocampal CA1 area of rats in each group,and electron microscopy was used to observe the synaptic ultrastruc⁃ture of hippocampal CA1 area.[Results]Ultrastructure of synapse in hippocampal CA1 area of model rats:There were no significant differences in the synaptic gap among groups(P>0.05);The PSD thickness,the curvature of the synaptic interface and the length of synaptic active zone were increased in rats significantly in Huangpu Tongqiao group than those in the model group(P<0.01),and there was no statistically significant difference in Huangpu Tongq⁃iao group compared with those in the Donepezil group(P>0.05).[Conclusion]Huangpu Tongqiao capsule can im⁃prove the synaptic structure plasticity of hippocampal CA1 area.
