摘 要:目的观察茵陈草水提物对四氯化碳致大鼠肝纤维化的干预作用。方法通过向SD大鼠腹腔注射四氯化碳与花生油混合溶液诱导建立肝纤维化大鼠模型。将50只肝纤维化模型大鼠按照随机数字表法随机分为五组:模型组、阳性药水飞蓟素40mg/kg组、茵陈草水提物2.5、5、10g/kg剂量组,每组10只。连续灌胃给药56天,每天1次。另设10只SD大鼠为正常组。模型组、正常组大鼠每天给予等体积生理盐水灌胃。末次给药4h后,3%苯巴比妥钠腹腔注射麻醉后,腹主动脉取血,离心提取血清。剥离肝脏,HE染色法及Masson染色法观察大鼠肝脏病理和纤维化程度。比色法检测大鼠血清天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)含量,ELISA法检测肝组织肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)含量。比色法检测肝组织透明质酸(HA),Ⅲ型前胶原(PCⅢ),层粘连蛋白(LN)、IV型胶原(IV-C)含量。结果与模型组比较,茵陈草水提物低、中、高剂量组大鼠血清AST、ALT水平显著降低[AST:(31.78±3.29)U/mL、(24.85±2.23)U/mL、(15.94±2.27)U/mL比(38.98±4.71)U/mL;ALT:(29.75±2.48)U/mL、(22.37±2.15)U/mL、(16.63±2.75)U/mL比(34.21±4.79)U/mL,P均<0.05],肝组织炎症因子TNF-α、IL-1β水平显著降低[TNF-α:(27.54±2.92)pg/mL、(23.27±2.56)pg/mL、(17.08±2.15)pg/mL比(31.54±2.81);IL-1β:(35.41±2.41)pg/mL、(27.26±1.37)pg/mL、(19.57±1.69)pg/mL比(42.63±3.53)pg/mL,P均<0.05];茵陈草水提物低、中、高剂量组大鼠肝组织胶原HA、PCⅢ、LN、IV-C水平显著降低[HA:(202.83±14.71)μg/L、(137.05±10.24)μg/L、(96.47±7.48)μg/L比(252.42±16.85)μg/L;PCⅢ:(171.87±8.23)μg/L、(114.75±11.35)μg/L、(68.98±7.83)μg/L比(191.76±14.53)μg/L;LN:(281.82±19.51)μg/L、(178.21±16.39)μg/L、(94.92±8.54)μg/L比(341.28±17.69)μg/L;IV-C:(25.52±2.42)μg/L、(21.85±2.26)μg/L、(18.37±2.48)μg/L比(28.77±3.29)μg/L,P均<0.05];茵陈草水提物高剂量组作用优于低、中剂�Objective To observe the effect of aqueous extract of Artemisia dracunculus L.on carbon tetrachloride-induced liver fibrosis in rats.Methods SD rats were intraperitoneally injected with mixed solution of carbon tetrachloride and peanut oil to established liver fibrosis models.Liver fibrosis rats were randomly divided into 5 groups with 10 animals per group:model control group,silymarin positive control group(40 mg/kg),Artemisia dracunculus L.aqueous extract groups(2.5,5 and 10 g/kg).Animals were intragastric administration for 56 days,once a day.Ten healthy SD rats were considered as normal control.Model and normal control rats were given equal volume saline,respectively.Four hours after last administration,rats were intraperitoneal anesthesia by 3%phenobarbital sodium.Blood from abdominal aorta was taken and serum was collected by centrifugation.Liver samples were collected for HE and Masson staining to observe pathological alterations and fibrosis.Serum aspartate amino-transferase(AST)and alanine aminotransferase(ALT)were determined.Inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in liver tissues were detected by ELISA.Hyaluronic acid(HA),type Ⅲ procollagen(PCⅢ),laminin(LN)and type IV collagen(IV-C)were detected by colorimetry.Results Compared to model control,low,medium and high dose group of Artemisia dracunculus L.aqueous extract decreased the serum levels of AST[(31.78±3.29),(24.85±2.23)and(15.94±2.27)vs(38.98±4.71)U/mL],and ALT[(29.75±2.48),(22.37±2.15)and(16.63±2.75)vs(34.21±4.79)U/mL],P<0.05,respectively;And also inflammatory factors TNF-α[(27.54±2.92),(23.27±2.56)and(17.08±2.15)vs(31.54±2.81)pg/mL],and IL-1β[(35.41±2.41),(27.26±1.37)and(19.57±1.69)vs(42.63±3.53)pg/mL]in liver fibrosis,as well as the liver collagen HA[(202.83±14.71),(137.05±10.24)and(96.47±7.48)vs(252.42±16.85)μg/L],PCⅢ[(171.87±8.23),(114.75±11.35)and(68.98±7.83)vs(191.76±14.53)μg/L],LN[(281.82±19.51),(178.21±16.39)and(94.92±8.54)vs(341.28±17.69)μg/L],and IV-C[(25.52�
