摘 要:利用16个表达序列标签简单重复序列(EST-SSR)标记对日本落叶松种子园遗传多样性进行分析评价,旨在明确该种子园的遗传基础,为后期育种策略的制定和遗传改良的可持续发展提供参考依据。结果表明:群体平均有效等位基因数(Ne)为2.542,平均观测杂合度(Ho)为0.578,平均期望杂合度(He)为0.528,平均Shannon多样性指数为0.972,平均多态信息含量指数(PIC)为0.485,日本落叶松种子园遗传多样性较高;群体中大部分位点均符合哈迪-温伯格平衡(HWE),群体内近交系数(Fis)、群体总近交系数(Fit)均为负值,群体表现为轻微杂合子过量,不存在近交衰退;不同来源的5个群体遗传多样性各参数值差异不大,群体间遗传距离计算、分子方差分析和主坐标分析结果均表明群体间遗传分化非常小,主要原因在于国内各栽培区引种交流频繁。Sixteen expressed sequence tag simple sequence repeat markers were used to analyze and evaluate the genetic diversity of Larix kaempferi in seed orchards,clarify the genetic basis of seed orchards,and provide a basis for the formulation of further breeding strategies and the sustainable development of genetic improvement.The results showed that in the population the average number of effective alleles(Ne)was 2.542,the average observed heterozygosity(Ho)was 0.578,the average expected heterozygosity(He)was 0.528,the average Shannon diversity index was 0.972,the average polymorphic information content index(PIC)was 0.485,and the genetic diversity of L.kaempferi seed orchards was high.Most of the loci in the population were consistent with HWE,and the inbreeding coefficient lines Fis and Fit were both negative.The population showed a slight heterozygote overabundance and no inbreeding recession.The results of genetic distance calculation,molecular variance analysis,as well as principal coordinates analysis indicated that the genetic differentiation between the populations was very small,mainly because of the frequent exchange of introduced species in different cultivation areas in China.
