摘 要:目的探讨杜仲黄酮对糖尿病肾病(DN)小鼠肾脏氧化应激及其Nrf2/HO-1信号通路的影响。方法 昆明种小鼠经静脉注射链脲佐菌素诱导糖尿病肾病模型,成功诱导的DN小鼠随机分组为模型对照组、缬沙坦对照组、杜仲黄酮低、高剂量组[80、160 mg/(kg·d)],并以健康昆明小鼠为正常对照。各组小鼠灌胃给药连续60 d,末次给药后收集小鼠血清和肾组织标本用于检测空腹血糖、肾功能指标、氧化应激相关指标和核转录因子E2相关因子2(Nrf2)/血红素氧合酶1(HO-1)信号通路蛋白表达。结果与模型组比较,杜仲黄酮给药组空腹血糖有不同程度下降,肾功能指标也明显改善。此外,杜仲黄酮组肾脏组织中超氧化物歧化酶活性升高,而丙二醛下降,Nrf2和HO-1蛋白表达升高(P<0.05)。结论杜仲黄酮对糖尿病肾病小鼠具有降低空腹血糖和改善肾功能作用,其作用机制可能与其能通过调节Nrf2/HO-1信号通路,进而改善肾脏组织氧化应激状态有关。Objective To investigate the effect of Eucommia flavonoids on renal oxidative stress and Nrf2/HO-1 signaling pathway in diabetic nephropathy(DN)mice.Methods Kunming mice were injected with streptozotocin to establish DN model.Subsequently,the DN mice were divided into following groups:model control group,valsartan control group,Eucommia flavonoids groups[80 mg/(kg·d)and 160 mg/(kg·d)],and healthy Kunming mice were considered as normal control group.The mice were administered drugs for 60 days,the serum and kidney samples were collected after last administration for fasting blood glucose,renal functional index and oxidative stress related indicators detections.The expressions of nuclear transcription factor E2 related factor 2/heme oxygenase 1(Nrf2/HO-1)signaling pathway proteins were also determined.Results Compared with the model group,the fasting blood glucose of the Eucommia flavonoids group decreased to varying degrees,and the renal function index also improved significantly.In addition,the activity of superoxide dismutase in the kidney tissue of the Eucommia flavonoid group increased,while the content of malondialdehyde decreased,and the expression of Nrf2 and HO-1 protein increased(P<0.05).Conclusion Eucommia flavonoids can reduce fasting blood glucose and improve renal function in diabetic nephropathy mice,and its mechanism may be that it can improve the oxidative stress of kidney tissues by regulating the Nrf2/HO-1 signaling pathway.
