摘  要:目的研究利拉鲁肽对高糖诱导人脐静脉内皮细胞(HUVECs)损伤的抗氧化作用及相关机制。方法分离培养HUVECs细胞,分为:N组(对照组)、G组(高糖组)、A组(低剂量高糖组)、B组(高剂量高糖组),各组均干预12、24、48 h后,测定其氧化应激产物和抗氧化产物含量。结果G组氧化应激产物活性氧簇(ROS)、丙二醛(MDA)、氧化低密度脂蛋白(ox-LDL)及Nrf2、血红素氧合酶1(HO-1)、醌氧化还原酶1(NQO1)、谷胱甘肽过氧化物酶(GSH-Px)的mRNA表达均明显高于N组(P<0.05);利拉鲁肽干预组细胞中的ROS、MDA、ox-LDL含量均明显低于G组(P<0.05);利拉鲁肽干预组细胞中Nrf2、NQO1、HO1、GSH-PX的mRNA表达量明显高于高糖组(P<0.05);利拉鲁肽的作用效果与干预时间和利拉鲁肽浓度呈正相关。结论利拉鲁肽可通过增强高糖条件下内皮细胞抗氧化因子的活性,减少氧化物生成,减轻内皮细胞氧化损伤。Objective To study the anti-oxidation effects and related mechanisms of liraglutide on high glucose-induced injury of human umbilical vein endothelial cells (HUVECs).Methods HUVECs cells were isolated and cultured, and were divided into group N (control group), group G (high-glucose group), group A (low dose high-glucose group) and group B (high dose high-glucose group). After 12, 24 and 48 h of intervention in each group, the contents of oxidative stress products and antioxidant products were measured.Results The contents of oxidative stress products of reactive oxygen species (ROS), malondialdehyde (MDA) and oxidized low density lipoprotein (ox-LDL) and mRNA expression levels of Nrf2, heme oxygenase 1 (HO-1), quinone oxidoreductase 1 (NQO1) and glutathione peroxidase (GSH-Px) in high-glucose group were significantly higher than those in control group (P<0.05). The contents of ROS, MDA and ox-LDL in cells in liraglutide intervention group were significantly lower than those in high-glucose group (P<0.05). The mRNA expression levels of Nrf2, NQO1, HO1 and GSH-PX in cells in liraglutide intervention group were significantly higher than those in high-glucose group (P<0.05). The liraglutide effects were positively correlated with intervention time and liraglutide concentration.Conclusions Liraglutide can reduce the formation of oxides and relieve the oxidative damage of endothelial cells by enhancing the activities of endothelial antioxidant factors under high glucose conditions.
