摘  要:目的:探讨黄芪多糖对2型糖尿病(T2DM)患者内皮祖细胞(EPCs)归巢能力的影响方法:培养、鉴定EPCs;建立裸鼠颈动脉损伤模型;PKH26标记的EPCs;分四组:健康人组、黄芪多糖干预组、糖尿病组(即未干预组)、PBS组,前三组经尾静脉移植相应的PKH26已标记的EPCs,PBS组尾静脉注射PBS液;于3和7d后取四组裸鼠损伤动脉,冰冻切片荧光显微镜下观察移植的细胞在内膜处的表达;纵行剖开后荧光显微镜下观察移植的EPCs定向归巢到血管损伤部位的情况并计数分析。结果:同一时间内,2型糖尿病患者EPCs归巢能力较健康人明显降低(P<0.01);2型糖尿病患者EPCs经黄芪多糖干预后,其归巢能力较未干预的糖尿病人增强(P<0.05)。结论:糖尿病患者EPCs在体内归巢的能力下降;糖尿病患者EPCs给予黄芪多糖干预后能增强其定向归巢到血管损伤部位的能力。Objective:To investigate the effect of Astragalus polysaccharides on the homing ability of endothelial progenitor cells(EPCs)in patients with type 2 diabetes mellitus(T2DM)Methods:EPCs were cultured and identified.PKH26-labeled EPCs were established in nude mice.They were divided into four groups:healthy person group,astragalus polysaccharide intervention group,diabetes group(ie,no intervention group),PBS group,and the first three groups.The corresponding PKH26 labeled EPCs were transplanted,and the PBS group was injected with PBS in the tail vein.After 3 and 7 days,the injured arteries of the four groups of nude mice were taken,and the expression of the transplanted cells in the inner membrane was observed under frozen microscope.Post-fluorescence microscopy was used to observe the homing of transplanted EPCs to the site of vascular injury and to count the analysis.Results:At the same time,the homing ability of EPCs in type 2 diabetic patients was significantly lower than that in healthy controls(P<0.01).The homing ability of EPCs in type 2 diabetic patients after intervention with Astragalus polysaccharides was enhanced compared with uninterrupted diabetics(P<0.05).Conclusion:The ability of EPCs to homing in the body of diabetic patients is decreased.The EPCs in diabetic patients can enhance the ability of directional homing to vascular injury sites after intervention with Astragalus polysaccharides.
