摘  要:目的探讨EGB761注射液对脉冲噪声性耳聋大鼠听功能的保护作用及其作用机制。方法36只雄性SD大鼠,空白对照组6只不予任何处理,其余30只同一脉冲噪声暴露后,随机分为3组:单纯噪声组、噪声+生理盐水组和噪声+EGB761组,每组各10只。噪声+EGB761组大鼠给予腹腔注射EGB761(3mL/kg,2/日,连用7日),噪声+生理盐水组注射等量生理盐水。采用听性脑干反应(ABR)检测各组大鼠听力情况,实时定量PCR法检测耳蜗中miR-183表达,HE染色研究耳蜗病理改变。结果单纯噪声组、噪声+生理盐水组、噪声+EGB761组大鼠ABR阈值均显著高于对照组(P<0.05);噪声+EGB761组大鼠ABR阈值明显低于单纯噪声组和噪声+盐水组(P<0.05)。单纯噪声组和噪声+生理盐水组均存在耳蜗渗血改变,而空白组和噪声+EGB761组耳蜗未见明显血性渗出及结构损坏。实时定量PCR结果显示,给予脉冲噪声刺激后,miR-183相对表达量较刺激前下降(P<0.05),EGB761治疗后相对表达量呈上升趋势(P<0.05)。结论EGB761可以减少脉冲噪声对耳蜗的损伤,上调miR-183表达,对耳蜗具有保护作用。Objective To investigate the protective effect of EGB761 on hearing function of rats with impulse noise induced deafness and its possible mechanisms.Methods Thirty-six male SD rats were selected and 6 served as the blank control group without any treatment.The remaining 30 rats were exposed to the same impulse noise and randomly divided into three groups:the noise group,the noise+saline group and the noise+EGB761 group,with 10 rats in each group.The noise+EGB761 group was given intraperitoneal injection of EGB761(3 mL/kg,2/day,for 7 days),and the noise+saline group was given intraperitoneal injection of saline.After the experiment,auditory brainstem response(ABR)was used to detect the hearing status of rats in each group,the expression of miR-183 in the cochlea was detected by quantitative real-time PCR,and the pathological changes of the cochlea were studied by HE staining.Results The ABR thresholds of rats in the noise group,the noise+saline group,the noise+EGB761 group were higher than those in control group(P<0.05);the ABR thresholds of rats in noise+EGB761 group were lower than those in noise group and noise+saline group(P<0.05).HE staining of the cochlea showed no obvious hemorrhagic exudation and structural damage in the control group and noise+EGB761 group,while there was hemorrhagic change in the noise group and noise+saline group.Real-time PCR results showed that the relative expression of miR-183 decreased after pulsed noise stimulation(P<0.01),and increased significantly after treatment with EGB761(P<0.05).Conclusion EGB761 can reduce the damage of cochlea caused by impulse noise,up-regulate the expression of miR-183 and protect the cochlea of rats.
