摘  要:为了获得准确的血凝(HA)试验结果,采用新城疫诊断技术(GB/T 16550-2008)中HA试验规范操作方法,检测抗凝血存放的容器、4℃条件下1%红细胞悬液的保存时间、LaSota保存温度和冻融次数、稀释液的种类等对HA结果的影响。结果显示:5 mL抗凝血在100 mL烧杯内于4℃存放120 h、1%红细胞悬液在4℃保存24 h、LaSota尿囊液在4℃条件下存放72 h、冻融不超过2次,HA结果均稳定无变化,PBS的缓冲能力比0.9%氯化钠强,此外在含有2%葡萄糖的0.75%氯化钠溶液作为稀释液时,可在48 h内维持HA效价均稳定无变化。In order to obtain accurate results of the hemagglutination test(HA test),the standard operation method of the HA test in the Newcastle disease diagnosis technique(GB/T 16550-2008)was adopted;the factors affecting the HA test concerning the storage vessel of anticoagulation blood,the storage time of 1%erythrocyte suspension at 4℃,the storage temperature,the times of freeze-thawing and the type of buffer were tested in this study.The results showed that the results of HA were stable and unchanged with 5 mL anticoagulant blood stored in a 100 mL beaker at 4℃for 120 h,1%erythrocyte suspension at 4℃for 24 h,LaSota allantoic fluid stored at 4℃for 72 h,freeze-thawing for no more than twice.The buffering capacity of PBS was stronger than that of 0.9%sodium chloride.In addition,the 0.75%sodium chloride solution containing 2%glucose could maintain the stability of HA titer within 48 h.
