摘  要:目前中草药防控肉鸡脂肪肝出血综合征(FLHS)缺乏确切理论依据,为了深入探讨茵陈提取物对FLHS的相应作用机制,研究不同剂量茵陈提取物对肉鸡PPARγ、NF-κB表达和相关血清细胞因子的影响。100只7日龄AA肉雏鸡随机分为5组:对照组、FLHS模型组及茵陈提取物低、中、高剂量组。采用索氏化学抽提法测定肝脂率、腹脂重/活重和肝脏出血指数,HE染色法观察肉鸡肝组织细胞,并统计FLHS发病率;ELISA试剂盒测定肉鸡血清TNF-α、IL-6血清含量,qRT-PCR方法检测肉鸡脂肪PPARγ、肝脏NF-κB mRNA相对表达量。结果显示:茵陈提取物可使肉鸡肝脂率、腹脂重/活重、肝脏出血指数显著下降(P<0.05或P<0.01),变性、出血肝细胞区域和数量显著减少,鸡血清TNF-α、IL-6含量以及NF-κB mRNA和PPARγmRNA表达水平显著下降(P<0.05或P<0.01)。研究表明,茵陈提取物可能通过抑制NF-κB/TNF-α反馈通路和PPAR-γmRNA表达,显著抑制TNF-α、IL-6等细胞因子含量,达到降低肝细胞脂肪变性,减少腹脂沉积及FLHS发病率的效果。The prevention and control of fatty liver hemorrhagic syndrome(FLHS)in broilers by Chinese herbal medicine lacks a definite theoretical basis.In order to further explore the mechanism of Artemisia capillaris extract on FLHS in broilers,the effect of different doses of Artemisia capillaris extract on the expression of PPARγ,NF-κB genes and related serum cytokines in broilers were observed.One hundred 7-day-age AA broilers were randomly divided into control group,FLHS model group,Artemisia capillaris extract low,medium and high dose group.The liver fat rate,abdominal fat weight to body weight ratio and hepatic hemorrhage index were measured by Soxhlet chemical extraction.The liver cells of broilers were observed by HE staining.FLHS incidence was calculated,serum TNF-αand IL-6 were detected by ELISA kit,and the relative expression levels of PPARγin adipose tissue and NF-κB mRNA in broiler liver were detected by qRT-PCR.The results showed that the liver fat rate,abdominal fat weight to live weight ratio,hepatic hemorrhage index,the area and number of degeneration,number of hemorrhagic hepatocytes,serum concentrations of TNF-αand IL-6,relative expression level of NF-κB mRNA and PPARγmRNA of Artemisia capillaris extract groups were decreased significantly(P<0.05 or P<0.01).It′s indicated that Artemisia capillaris extract may inhibit the NF-κB/TNF-αfeedback pathway and PPAR-γmRNA expression,decrease the serum concentrations of TNF-αand IL-6,relieve the degeneration of liver cells,reduce abdominal fat deposition and incidence of FLHS.
