摘  要:目的 :观察人脐带间充质干细胞(human umbilical cord-derived mesenchymal stem cells, hUC-MSCs)膝关节内注射对大鼠的毒性反应。方法 :实验分为生理盐水对照组、有血清MSCs低剂量组和高剂量组、无血清MSCs低剂量组和高剂量组。观察大鼠一般生命体征和血液尿液指标;取主要脏器组织进行病理学检查。结果 :注射MSCs后,MSCs组与生理盐水对照组相比,大鼠一般生命体征、血常规、T淋巴细胞亚群、血清细胞因子、尿常规,均无显著差异;主要脏器组织病理学无明显改变。一部分血液生化指标虽然在MSCs注射后有所波动,但仍在参考范围内或接近参考范围上限。结论 :膝关节内注射hUC-MSCs对大鼠没有明显的毒性反应。Objective:To investigate the toxic reactions of intra-articular administration of human umbilical cordderived mesenchymal stem cells(hUC-MSCs)in rats.Methods:Sprague-Dawley(SD)rats were randomly divided into a normal saline control group,a low-dose F-MSCs(MSCs cultured with fetal bovine serum)group,a high-dose F-MSCs group,a low-dose SF-MSCs(MSCs cultured with serum-free medium)group,and a high-dose SF-MSCs group.General vital signs,blood routine,blood biochemistry,subsets of T cells,serum cytokines,routine urine,pathology of vital organs and tissues were respectively determined.Results:There were no obvious differences between the MSCs group and the control group in all other indicators except some blood biochemistry indicators.Conclusion:There was no obvious toxic effect of intra-articular administration of hUC-MSCs on rats.
