摘 要:目的:观察雷公藤多苷(TWP)对角叉菜胶诱导的慢性前列腺炎大鼠模型的抗增生作用并探讨其可能的作用机制。方法:30只SD大鼠分为正常对照组、模型组和TWP组,每组10只,采用50μL 3%角叉菜胶注入SD大鼠前列腺双侧腹叶建立慢性前列腺炎模型,连续给药4周。通过病理组织切片评价TWP对前列腺炎模型大鼠的抗增生和抗炎作用,免疫组化观察波形蛋白(vimentin)和钙粘蛋白(e-cadherin)表达,Western blot检测转化生长因子-β1(TGF-β1)、转化生长因子-β2(TGF-β2)和环氧合酶(COX-2)蛋白表达,ELISA法测定前列腺组织及血浆胰岛素样生长因子1(IGF-1)水平。结果:病理学观察可见,与正常对照组相比,模型组大鼠前列腺肿大,间质室面积增大,而TWP组较模型组显著减轻(P<0.05)。免疫组化检测结果显示,与正常对照组相比,模型组大鼠前列腺组织vimentin表达显著增加,而TWP组较模型组显著减少(P<0.05),而对于e-cadherin表达无显著影响。Western blot检测结果显示,与正常对照组相比,模型组大鼠前列腺组织TGF-β1、COX-2蛋白表达显著升高,TWP组较模型组显著降低(P<0.05)。ELISA检测结果可见与正常对照组相比,模型组大鼠列腺组织IGF-1水平升高,TWP组较模型组显著降低(P<0.05)。结论:TWP可能通过抑制组织的炎症反应并降低COX-2、TGF-β1、IGF1表达水平改善大鼠良性前列腺增生。Objective:To observe the anti-proliferative effect of Tripterygium Wilfordii(TWP)on rat model of chronic prostatitis induced by carrageenan and to explore its possible mechanism.Methods:Thirty SD rats were divided into normal control group,model group and TWP group,10 rats in each group,and 50μL of 3% carrageenan was injected into bilateral abdominal lobe of rats’prostates for 4 weeks to establish a model of chronic prostatitis.The pathological tissue sections were used to evaluate the anti-proliferative and anti-inflammatory effects of TWP on prostatitis model rats.The positive staining area of vimentin and e-cadherin was observed by immunohistochemistry.The expressions of transforming growth factor-β1(TGF-β1),TGF-β2 and cyclooxygenase(COX-2)protein were detected by Western blot.Insulin-like growth factor 1(IGF-1)levels in the lateral prostate tissue and plasma were determined by ELISA.Results:Pathological observation showed that compared with the normal control group,the rats in the model group had enlarged prostates and the area of the interstitial compartment increased,while the symptoms listed above in the TWP group were significantly reduced compared with the model group(P<0.05).The results of immunohistochemical examination showed that compared with the control group,vimentin increased in the model group,while the TWP group was significantly reduced(P<0.05),without significant effect on e-cadherin.Western blot results showed that compared with the normal control group,the expression of TGF-β1 and COX-2 protein in the prostate tissue of the model group was significantly increased,and the TWP group was significantly lower than the model group(P<0.05).The ELISA test results showed that compared with the normal control group,the IGF-1 level in the rat glandular tissue of the model group was increased,and the TWP group was significantly lower than the model group(P<0.05).Conclusion:The anti-proliferative effect of TWP on prostatitis rats may be through inhibiting the inflammatory response of tissues and
