摘 要:目的观察弥漫性脑损伤(diffuse brain injury,DBI)后不同时间海马区Wnt3a、β-catenin、CyclinD1的表达,探讨Wnt/β-catenin信号通路在脑损伤中的作用机制及与损伤的时间关系。方法参照Marmarou原理建立DBI模型,将大鼠分为正常对照组及DBI 6h、1d、2d、5d、7d组,应用免疫组化染色及蛋白免疫印迹法观察Wnt3a、β-catenin、CyclinD1的表达及变化。结果正常对照组大鼠海马区Wnt3a、β-catenin、CyclinD1均有少量表达,与正常对照组相比,模型组海马区Wnt3a、β-catenin在伤后6h升高,1d达到高峰,Wnt3a在伤后7d再呈升高趋势,β-catenin在伤后7d降至正常水平;CyclinD1在伤后6h达到高峰,ld^7d持续降至正常水平,差异均有统计学意义(P<0.05)。结论Wnt/β-catenin信号通路在弥漫性脑损伤中具有重要调控作用,该信号Wnt3a、β-catenin、CyclinD1蛋白表达在时间上存在着差异性,对弥漫性脑损伤分子病理机制的研究及损伤时间推断具有一定的指导意义。Objective To observe the expression of Wnt3a,β-catenin and CyclinD1 in hippocampus at different time after diffuse brain injury,and to explore the mechanism of Wnt/β-catenin signaling pathway in brain injury and its relationship with the time of injury.Methods DBI model was established according to Marmarou principle.Rats were divided into normal control group and DBI 6h,1d,2d,5d and 7d groups.The expression and changes of Wnt3a,β-catenin and CyclinD1 were observed by immunohistochemical staining and Western blot.Results The expression of Wnt3a,β-catenin and CyclinD1 in hippocampus of normal control group rats was slight.Compared with the normal control group,Wnt3a andβ-catenin in the hippocampus of the model group increased at 6h after injury,reached the peak at 1d,Wnt3a increased at 7d after injury,andβ-catenin decreased to normal at 7d after injury.CyclinD1 reached the peak at 6h after injury,and ld-7d continued to fall to normal levels.The differences were statistically significant(P<0.05).Conclusion Wnt/β-catenin signaling pathway plays an important regulatory role in diffuse brain injury.The expression of Wnt3a,β-catenin and CyclinD1 proteins is different in time,which has certain guiding significance for the study of molecular pathological mechanism of diffuse brain injury and the estimation of injury time.
