摘  要:通过对细胞活力、增殖、迁移、细胞周期、胞内活性氧(ROS)和抗氧化能力的检测,分析条纹斑竹鲨(Chiloscyllium plagiosum)鱼皮多肽对人永生化表皮角质形成细胞(HaCAT细胞)的作用.实验结果显示:随着多肽作用浓度的增加,HaCAT细胞活力显著提升,最高达144%;细胞单克隆增殖能力明显增强;细胞划痕迁移愈合速率增加;细胞周期无显著变化;胞内ROS水平显著降低,最低达49.1%;在H2O2氧化损伤下细胞活力明显恢复,最高达119%.上述结果表明条纹斑竹鲨鱼皮多肽对HaCAT细胞的多项生理生化活性具有促进作用,可为其作为功能性皮肤敷料的开发研究提供参考.In this study,effects of Chiloscyllium plagiosum skin peptides on human immortalized keratinocytes(HaCAT cells)were investigated in the aspect of cell activity,proliferation,migration,cell cycle,intracellular reactive oxygen species(ROS)and oxidative protection.The results showed that the activity of HaCAT cells increased significantly after exposure to the peptides,and the maximum activity increased up to 144%.Moreover,the ability of cell monoclonal proliferation significantly enhanced,and the wound healing rate of cell scratches increased.There was no significant change in the cell cycle.Intracellular ROS level decreased significantly,with the minimum as low as 49.1%.Under the oxidative damage of H2O2,the cell activity was significantly restored with the peptide treatment,with the activity increasing up to 119%.These results demonstrate that C.plagiosum skin peptides could promote multiple physiological and biochemical activities of HaCAT cells,which may provide reference for the application study as functional skin dressings.
