摘 要:目的开发一种利用制备液相色谱系统从槐花米中制备高纯度芦丁的快速方法。方法槐花米粉末先用甲醇提取得到粗芦丁,粗芦丁再利用制备液相色谱系统分离纯化。结果使用SunFireRPrep C18 OBDTM柱(5μm,19 mm×250 mm),用0.2%乙酸水溶液-甲醇(45∶55,V/V)流动相等度洗脱,流速10 mL/min,检测波长360 nm,柱温为室温,进样量2.5 mL,采用核磁共振波谱技术对纯化芦丁进行鉴定。结论在该色谱条件下,可以有效从槐花米中分离制备芦丁,其核磁共振波谱图符合芦丁结构特征。Objective This work aims to develop a rapid method for the separation of rutin from Flos Sophorae Immaturus using the preparative HPLC.Methods The crude rutin was extracted from Flos Sophorae Immaturus by methanol,and then separated and purified using the preparative HPLC.Results A SunFireRPrep C18 OBDTM column(5μm,19 mm×250 mm)was used.The column was isocratic eluted with 0.2%aqueous acetic acid-methanol(45∶55,V/V).The flow rate was 10 mL/min.The detection wavelength was 360 nm.The column temperature was room temperature,and the injection volume was 2.5 mL.The purified rutin was identified by NMR spectroscopy.Conclusion Rutin can be separated effectively from Flos Sophorae Immaturus under this chromatographic condition,and its NMR spectra conform to the structural characteristics of rutin.
