摘　　要：目的探究髓样锌指1(MZF1)对MMP-2的影响及其在宫颈癌细胞迁移和侵袭中的作用。方法使用Jet-PEI试剂,将pcDNA3.0(Neo)或pcDNA-MZF1(MZF1)转染细胞,培养过表达Neo或MZF1的SiHa细胞,构建稳定转染的细胞系;通过Western blot分析、RT-PCR实验、细胞活性检测、细胞周期分布、明胶酶谱法、细胞迁移和侵袭等实验论证过度表达的MZF1对MMP-2的影响,进而分析MZF1对宫颈癌细胞迁移和侵袭的作用。结果过表达MZF1的SiHa细胞运动减少了70%,穿过Matrigel基质胶的细胞数量比转染Neo的细胞减少了80%(P<0.05);Western blot、明胶酶谱法以及RT-PCR实验结果证明:MZF1过表达抑制了MMP-2活性(P<0.05),对MMP-9活性不产生影响(P>0.05)。结论 MZF1过度表达可抑制人宫颈癌SiHa细胞的迁移和侵袭,并涉及MMP-2蛋白活性及基因表达。Objective To investigate the effect of myeloid zinc finger protein 1(MZF1)overexpression on MMP-2 protein and gene levels in human cervical cancer cells,and the migration and invasion of human cervical cancer cells. Methods Stable SiHa cells overexpressing MZF1 was established: SiHa cells were transfected with pcDNA3.0(Neo)or pcDNA-MZF1(MZF1)using Jet-PEI reagent according to manufacturer’s instructions.SiHa cells overexpressing Neo or MZF1 were cultured,then the effect of overexpressing MZF1 on MMP-2 level were demonstrated by a series of experiments such as Western blot,cell survival assay,cell cycle analysis,gelatin zymography and cell migration and invasion experiments. Its influence on human cervical cancer cells migration and invasion was further analyzed. Results Migration and invasion experiments showed that the migration ability of SiHa cells overexpressing MZF1 was reduced by a 70% decrease in cell movement,the number of cells overexpressing MZF1 that moved through the membrane of the Matrigel chamber was about 80%less than that of Neo cells(P<0.05). Western blot,gelatin zymography and RT-RCR results showed that the overexpression of MZF1 reduced MMP-2 activity in protein and m RNA levels(P<0.05),without affecting MMP-9 activity(P>0.05). Conclusion Overexpression of MZF1 can inhibit the migration and invasion of human cervical cancer SiHa cells,involving the activity of MMP-2 protein and gene expression.
