骨刺散对大鼠佐剂关节炎模型继发性病变的影响

摘　　要：目的:探讨骨刺散对大鼠佐剂关节炎模型继发性病变足肿胀及体质量和脏器系数的影响。方法:取80只Wistar大鼠随机分为空白对照组,模型对照组,阳性预防组,阳性治疗组(0.5 mL/只),大、小剂量骨刺散白醋预防组,大、小剂量骨刺散白醋治疗组(0.6 g/只,0.3 g/只)。除空白对照组外其余各组造模,在右后足跖皮内注射弗氏完全佐剂0.l mL致炎,制备佐剂诱导关节炎大鼠模型。注射佐剂当日即开始给药为阳性预防组,第8天开始给药为阳性治疗组,每日给药1次,到造模21天后,停药观察3天,第24天取血杀剖动物,秤取胸腺、脾质量。注射佐剂前及后3 h以及间隔一定日期各测左右足爪容积1次,每隔7天,测体质量1次。结果:造模后11天,模型对照组大鼠左后踝关节肿胀明显,于第17 d达到高峰。给予骨质宁搽剂和骨刺散后,大鼠左侧继发性肿胀有明显改善。与空白对照组比较,模型对照组大鼠体质量增长缓慢;与模型对照组比较,阳性药物组,骨刺散大、小剂量组大鼠体质量明显增长,但预防组相比治疗组效果要好。与空白对照组相比,模型对照组大鼠脾脏和胸腺指数显著下降,阳性药物组,大、小剂量骨刺散组脾脏和胸腺指数恢复正常。结论:骨刺散能明显抑制大鼠佐剂性关节炎致炎足肿胀、对大鼠体质量有一定的改善作用,明显增加大鼠胸腺、脾脏系数。Objective:To discuss the effects of Guci powder on paw swelling,body mass and organ coefficient in rat model suffering adjuvant arthritis.Methods:Eighty Wistar rats were randomized into blank control group,model control group,positive prevention group,positive treatment group(0.5 mL each rat),the prevention groups of high and small dose of Guci powder and white vinegar,and the treatment groups of high and small dose of Guci powder and white vinegar(0.6 g each rat,0.3 g each rat).Except blank control group,other groups were prepared into the models by injecting Freund's complete adjuvant,0.1 mL,into the right posterior plantar.The group that began to medicate at the injection day was positive prevention group,the group medicating at the eighth day of injection was positive treatment group,medicating once each day,at the 21st day of modelling,they were observed for three days after drug withdrawal,the rats were sacrificed at the 24th day to collect blood and weigh thymus and spleen.Claw volume of left and right paws were measured before,three hours after the injection,after a certain interval,the interval was seven days,to detect body mass each time.Results:Left posterior ankle of the rats in model control group became swollen obviously 11 days after modelling,reached the peak at the 17th day.After administering with Guzhining liniment and Guci powder,left secondary swelling of the rats has been improved notably.Compared with blank control group,body mass of the model group grew slowly;compared with the model group,body mass of the rats in large and small doses groups of Guci powder and positive medicine group increased remarkably,while the prevention group showed better effects compared with the treatment group.Compared with blank control group,spleen and thymus indexes of model control group reduced notably,while spleen and thymus indexes in positive medicine group,large and small doses groups of Guci powder restored to normal.Conclusion:Guci powder could notably inhibit paw swelling of the rats,it owns certain i
