摘  要:目的探讨石参提取物对鱼藤酮诱导的帕金森病体外模型的保护作用。方法以鱼藤酮诱导人神经母细胞瘤细胞SH-SY5Y细胞建立帕金森病体外模型,通过Cell Counting Kit-8法(CCK-8)研究药物的保护作用、2,7-二氯四氟乙烷双乙酸盐(DCFH-DA)法评估细胞内活性氧(ROS)、化学显色法测定细胞内丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)及谷胱甘肽过氧化物酶(GPx)的含量、Western blot法检测核因子E2相关因子2(Nrf2)及血红素加氧酶-1(HO-1)蛋白的表达量。结果石参提取物能有效对抗鱼藤酮引起的细胞损伤及细胞内活性氧升高,且能升高SOD、GSH及GPx含量同时降低MDA的含量,还能同时上调Nrf2及HO-1的蛋白表达量。结论石参提取物保护鱼藤酮诱导的SH-SY5Y细胞帕金森病模型的作用可能与增强Nrf2/HO-1通路功能有关。Objective To investigate the protective effect of the Uraria crinite extract on rotenone⁃induced Parkinson′s disease(PD)model in vitro.Methods The SH⁃SY5Y cells were induced to build a Parkinson′s disease model by rosterone,and the protective effect of drugs was studied by Cell Counting Kit⁃8(CCK⁃8),the intracellular reactive oxygen species(ROS)was evaluated by 2,7⁃dichlorotetrafluoroethane diacetate(DCFH⁃DA),the intra⁃cellular contents of malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH)and glutathione peroxidase(GPx)were determined by chemical coloration kit,and the expression level of nuclear factor E2⁃related factor 2(Nrf2)and heme oxygenase⁃1(HO⁃1)protein was detected by Western blot.Results The extract of Uraria crinite could effectively relieve the damage caused by rotenone,and reduced the intracellular ROS,increased the content of SOD,GSH and GPx,and reduced the content of MDA,and up⁃regulated the expression levels of Nrf2 and HO⁃1 protein.Conclusion The protective effect of Uraria crinite extracts on rotenone⁃induced cell model of PD may be related to the enhancement of Nrf2/HO⁃1 pathway function.
