摘 要:阿尔兹海默病(Alzheimer's disease,AD)是一种慢性大脑原发性神经系统退行性疾病,以记忆力减退、认知功能障碍和行为异常为主要特征,最终导致死亡。AD病因繁多,既有遗传基因突变的因素,更有后天生活方式和环境参与发病和疾病进展。其发病机制同样复杂,涉及多个环节,多途径以及多系统结构和功能的异常,目前其发病机制的学说有上十种,却没有一个能够全面准确地阐述AD的发病机制。随着对AD研究的不断深入,研发人员的研发理念从一药一靶向一药多靶转变,目前一药多靶的新颖药物实体的合理设计可能是对AD研究的最新趋势。本文讨论了近几年来发表的关于AD多靶点化合物的研究进展。以期后续研究能更好地了解AD的复杂性并确定有效的候选药物来彻底治愈这种破坏性疾病。Alzheimer's disease(AD)is an age-related neurodegenerative disorder of the brain,characterized by memory loss,cognitive impairment and decline in language skills,eventually lead to death.There are many causes of AD,including genetic mutations,acquired lifestyle and environment.The pathogenesis of AD is also complex,involving multiple links,pathways and abnormal structure and function of multiple systems.There are more than ten theories about its pathogenesis at present,but none of them can fully and accurately explain the pathogenesis of AD.With the development of AD research,the concept of R&D personnel has changed from"one drug one target"to"one drug multiple targets".In this paper,the research progress of multi-target compounds for Alzheimer's disease in recent years were reviewed and discussed.
