摘 要:目的研究并筛选达卡巴嗪脂质体的制备工艺及处方。方法分别采用逆向蒸发法和硫酸铵梯度法制备达卡巴嗪脂质体,以包封率为主要指标,通过单因素实验和正交实验优化处方。结果逆向蒸发法制备的达卡巴嗪脂质体最高包封率为(28.02±3.11)%,载药量为(2.71±0.32)%,硫酸铵梯度法制备的达卡巴嗪脂质体最高包封率为(63.67±2.04)%,载药量为(7.25±0.23)%。结论硫酸铵梯度法制备达卡巴嗪脂质体包封率高,载药量大,可作为达卡巴嗪脂质体的制备方法。Objective To study and optimize the preparation technology and formulation of dacarbazine(DTIC)liposomes.Methods DTIC liposomes were prepared by a reverse evaporation method and an ammonium sulfate gradient method,respectively.The preparation formulation was optimized by single factor experiments and orthogonal experiments.The drug entrapment efficiency was chosen as the main evaluating indicator of DTIC liposomes.Results The highest encapsulation efficiency and drug loading of DTIC liposomes prepared by the reverse evaporation was(28.02±3.11)%and(2.71±0.32)%,respectively.The highest encapsulation efficiency and drug loading of DTIC liposomes prepared by the ammonium sulfate was(63.67±2.04)%and(7.25±0.23)%,respectively.Conclusion Compared to the reverse evaporation method,the ammonium sulfate gradient method has advantages of higher drug entrapment efficiency and larger drug loading.It can be used as a preparation method for DTIC liposomes.
