摘 要:目的研究靳三针对脑卒中肢体痉挛大鼠肌张力增高的抑制作用及对钙结合蛋白小白蛋白(PV)、钙结合蛋白D(CB)表达的影响。方法将32只SD大鼠随机分为假手术组、模型组和靳三针组,采用线栓法制备脑卒中肢体痉挛大鼠模型,靳三针组于造模成功后第3日取穴治疗6 d,分别于术后1 d、3 d(治疗前)、9 d(治疗后)测定各组大鼠Bederson神经功能缺损评分及Feeney平衡木评分,采用电生理描记法测定术后3 d(治疗前)、9 d(治疗后)各组大鼠肌张力,采用免疫印迹(Western blotting)检测术后3 d(治疗前)、9 d(治疗后)各组大鼠脑干组织中PV、CB蛋白表达水平。结果与假手术组比较,模型组、靳三针组大鼠各时间点神经功能缺损评分、Feeney平衡木行走评分显著升高(P<0.05),间接肌张力及脑干组织PV、CB蛋白水平显著降低(P<0.05);术后9 d(治疗后),与模型组比较,靳三针组大鼠神经功能缺损评分、Feeney平衡木行走评分显著降低(P<0.05),间接肌张力及脑干组织中PV、CB表达水平显著升高(P<0.05);与术后3 d(治疗前)比较,术后9 d(治疗后)靳三针组大鼠神经功能缺损评分、Feeney平衡木行走评分显著降低(P<0.05),间接肌张力、脑干组织PV、CB蛋白水平显著升高(P<0.05)。结论靳三针可能通过上调钙结合蛋白PV、CB表达抑制脑卒中肢体痉挛大鼠肌张力增高,缓解其痉挛状态。Objective:To study the inhibitory effect of Jin′s 3-needle therapy on the increase of muscle tension and its influences on the expressions of PV and CB in rats with stroke limb spasm. Methods:32 SD rats were randomly divided into sham operated group,the model group and Jin′s 3-needle therapy group. The rat model of focal cerebral ischemia was established by thread occlusion. Jin′s 3-needle therapy group was treated with acupoints on the third day after the model was established successfully. On the 1 st,3 rd(before treatment)and 9 th day(after treatment)after operation,the scores of Bederson′s neurological deficit and Feeney balance beam walking score were measured;electrophysiological method was used to measure the muscle tension,and WB was used to detect the expressions of PV and CB proteins in brain stem tissue of rats before treatment and after treatment. Results:At different time points,compared with the sham operation group,the neurologic deficit score and Feeney balance beam walking score were significantly higher(P< 0.05),and the indirect muscle tension and the protein levels of PV and CB in brain stem of the model group and Jin′s 3-needle therapy group decreased significantly(P< 0.05). After treatment,compared with the model group,the neurological deficit score and Feeney balance beam walking score were significantly reduced(P< 0.05);the indirect muscle tension and the protein levels of PV and CB in brain stem of Jin′s 3-needle therapy group increased significantly(P< 0.05). Compared with before treatment,the neurological deficit score and Feeney balance beam walking score of rats in Jin′s 3-needle therapy group after treatment were significantly reduced(P< 0.05),and the indirect muscle tension and the protein levels of PV and CB in brain stem increased significantly(P< 0.05). Conclusion:Jin′s 3-needle therapy may inhibit the increase of muscle tension and relieve the spasticity by up-regulating the expression of calcium binding protein PV and CB.
