摘  要:目的:建立一种同时测定截叶铁扫帚中3个黄酮类成分(芦丁、槲皮素、山奈酚)含量的方法。方法:采用RP-HPLC法,ZORBAX Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm),甲醇-0.4%磷酸为流动相,检测波长371 nm,流速为0.7 mL/min,柱温35℃。结果:芦丁在0.108~1.681μg(r=0.9993)有良好的线性关系,槲皮素在0.204~4.083μg(r=0.9996)有良好的线性关系,山奈酚在0.072~1.440μg(r=0.9993)有良好的线性关系;平均加样回收率分别为99.37%(RSD=1.70%)、100.10%(RSD=1.80%)、99.42%(RSD=0.79%)。结论:建立的方法重复性好,精密度高,稳定性好,有很好的分离效果和较高的分析准确度,可用于截叶铁扫帚药材的质量控制。Objective:To establish a method for the simultaneous determination of rutin,quercetin and kaemphenol in Lespedeza cuneata.Methods:The content of rutin,quercetin and kaemphenol was determined by reversed phase high performance liquid chromatography(RP-HPLC).The chromatographic column was ZORBAX Eclipse XDB-C18 column(4.6 mm×250 mm,5μm).The mobile phase was methanol-0.4%phosphoric acid.The detection wavelength was 371 nm.The flow rate was 0.7 mL/min and the column temperature was kept at 35℃.Results:The linear ranges for rutin,quercetin and kaemphenol were 0.108-1.681μg(r=0.9993),0.204-4.083μg(r=0.9996),0.072-1.440μg(r=0.9993).The average recoveries for three flavonoids were 99.37%(RSD=1.70%),100.10%(RSD=1.80%),99.42%(RSD=0.79%),respectively.Conclusion:The method has a good reproducibility,high precision,good stability,good separation effect and high analytical accuracy,and can be used for the quality control of L.cuneata.
