彩叶桂新品种‘紫嫣公主’遗传多样性的AFLP和SSR分析

摘  要:以长叶木犀为对照,采用AFLP和SSR方法,研究了彩叶桂花(Osmanthus fragrans)新品种‘紫嫣公主’与其他木犀的遗传关系。筛选出55对AFLP引物和17个SSR,分别产生了1103个和45个扩增产物,其中多态性占AFLP的92.29%,占SSR的62.2%。在群体水平上,有效等位基因的数量从0.76到1.11不等,Shannon指数从0到0.11不等,表明桂花品种的遗传基础较窄。‘紫嫣公主’与其他桂花品种的遗传相似性在0.58~0.80。UPGMA聚类分析结果表明,‘紫嫣公主’属于银桂品种群。本研究结果可为桂花种质资源的保护和新品种选育提供新的分子依据。Changyemuxi was used as the control species.The genetic relationship between a new colorful Osmanthus fragrans cultivar“Ziyangongzhu”and other O.fragrans was determined by AFLP and SSR markers.Fifty-five AFLP primer pairs and 17 SSRs were screened,and 1103 and 45 amplification products were produced,of which 92.29%in AFLP and 62.2%in SSR were polymorphic.At the population level,the number of effective alleles varied from 0.76 to 1.11,and the Shannon index(I)ranged from 0 to 0.11,which indicated a narrow genetic base of O.fragrans cultivars.The genetic similarity varied from 0.61 to 0.8 between Ziyangongzhu and other O.fragrans cultivars.The UPGMA clustering revealed that Ziyangongzhu may belong to O.fragrans Albus group.The results can provide new molecular evidence for germplasm resources protection and breeding new cultivars.
