摘  要:多维高效液相色谱快速纯化物质是当前色谱领域一个重要的发展方向。本实验首次对全自动多维高效液相色谱系统进行了优化装配,并对草乌甲素(BLA)进行连续单柱三维的分离。从实验结果看出,本实验采用的在线单柱多维分离系统比普通的离线三维色谱分离纯化要快得多;因其在线的密闭系统实施全部操作,样品质量回收率更高。Multidimensional high performance liquid chromatography(MD-LC) system is a hot-point of research in the field of liquid chromatography. In this experiment, we used a fully-automatic MD-LC system firstly to separate and purify bulleyaconitine A(BLA) of ultra-high purity by 3 D-LC with a single reverse phase liquid chromatographic(RPLC) column. In comparing with conventional 3 D-LC by means of off-line manner, the online 3 D-LC system with the single RPLC column has some advantages of high speed, high mass recovery, due to all operations to be carried out in a closed system.
