摘  要:目的:明确扶正解毒方体内外对肺癌的抑制作用并探明其可能的作用机制。方法:体内实验:建立Lewis小鼠肺癌皮下移植瘤模型,不同浓度扶正解毒方灌胃干预,评价其对移植瘤增殖的影响;免疫组化检测Cleaved-caspase3的表达。体外实验:不同浓度扶正解毒方干预A549和H1299细胞,CCK8检测细胞活性,流式细胞仪检测凋亡情况。结果:扶正解毒方抑制Lewis小鼠肺癌皮下移植瘤增殖,促进移植瘤组织Cleaved-caspase3表达。CCK8结果显示扶正解毒方体外可明显抑制A549和H1299细胞株增殖活性,流式细胞仪检测提示扶正解毒方可促进肺癌细胞凋亡。结论:扶正解毒方体内、外有明确的抑制增殖和促凋亡作用,可能通过促进细胞凋亡发挥抑制肺癌增殖的作用。Objective:To study the inhibitory effect of the Fuzheng Jiedu prescription(扶正解毒方)on lung cancer,and to explore its possible mechanism of action.Methods:In vivo experiment,LLC-transplanted mouse models were established.They were given irrigation by different concentrations of the Fuzheng Jiedu prescription,to evaluate the inhibition on the transplanted tumor.The expression of Cleavedcaspase3 was detected by immunohistochemistry.In vitro experiment,A549 and H1299 cells were interfered by different concentrations of the Fuzheng Jiedu prescription,and cell proliferation activity was detected by CCK8,the intrinsic apoptosis was detected by flow cytometry.Results:The Fuzheng Jiedu prescription can inhibited the proliferation of the transplanted tumors and increased the expression of Cleavedcaspase3.The Fuzheng Jiedu prescription could significantly inhibit the proliferation of A549 and H1299 cells in vitro by CCK8 detected,and promote the apoptosis of lung cancer cells by flow cytometry.Conclusion:The Fuzheng Jiedu prescription shows a clear anti-tumor effect and pro-apoptotic effect in vivo and in vitro,and the inhibitory effect may be achieved by promoting apoptosis.
