摘  要:基于向量-子空间夹角判据,建立了淡水鱼中隐性孔雀石绿(LMG)的快速提取和紫外光谱检测方法。首先采用高效液相色谱-紫外可见光谱仪联用,建立不含待测组分LMG的淡水鱼本底数据库;然后采集LMG的标准光谱数据库,即可建立快速分析的数学模型。运用该模型,直接采集市售淡水鱼的紫外可见光谱即可快速计算LMG的含量。实验结果表明:快速分析法和高效液相色谱法结果接近,加标回收率在92~120%之间;乙腈提取法的提取率高于国标法,相对标准偏差小于5%,重现性好,解决了现有隐性孔雀石绿提取方法繁琐的问题;实现了淡水鱼中隐性孔雀石绿快速提取和检测。A rapid extraction and UV analytical method of LMG was established based on the criterion of vector-subspace.Firstly,the sample background spectrum database was constructed and separated by HPLC-UV,which does’t contain LMG spectra matrix deducted from the sample spectrum database.Secondly,collecting LMG standard spectrum.Then the mathematics model was constructed with the sample background matrix and LMG standard spectrum to determine LMG in fresh water fish by collecting the UV spectrum of sample.The study showed that the results of rapid analytical method were close to HPLC,the recovery rates were 92%~120%;the extraction rates with Acetonitrile were better than the national standard method,which the relative standard deviation of rapid pretreatment was within 5%with good reproducibility.This method was effective for rapid detection of LMG in fresh water fish and solved the problems of complex operation.
