摘 要:目的探讨氧化型低密度脂蛋白(oxidized low-density lipoprotein,ox-LDL)对小鼠主动脉内皮细胞中内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)的影响及可能机制。方法体外培养小鼠主动脉内皮细胞,按照0 mg/L组(对照1组)、25 mg/L组、50 mg/L组、100 mg/L组的ox-LDL浓度梯度分别处理24 h。Western blot检测eNOS、磷酸化eNOS和Caveolin-1(Cav-1)蛋白表达。为了进一步验证ox-LDL是通过影响Cav-1进而减弱eNOS和磷酸化eNOS蛋白表达,设立对照2组和siRNA组,应用Cav-1-mus小干扰RNA(siRNA)敲减Cav-1基因后,观察eNOS和磷酸化eNOS蛋白表达。结果与对照1组比较,25 mg/L组、50 mg/L组、100 mg/L组Cav-1、eNOS和磷酸化eNOS蛋白表达明显降低,差异有统计学意义(P<0.05,P<0.01)。siRNA组Cav-1、eNOS和磷酸化eNOS蛋白表达较对照2组明显降低(0.07±0.02 vs 0.45±0.12,P<0.01;0.34±0.13 vs 0.85±0.38,P<0.05;0.47±0.18 vs 0.86±0.13,P<0.05)。结论ox-LDL降低内皮细胞eNOS蛋白表达可能是通过影响Cav-1蛋白表达而发挥作用。Objective To study the effect of ox-LDL on endothelial nitric oxide synthase(eNOS)attenuation in aortic endothelial cells(AEC)from mice and its mechanism.Methods The AEC from mice were divided into 0 mg/L ox-LDL group(control group 1),25 mg/L ox-LDL group,50 mg/L ox-LDL group and 100 mg/L ox-LDL group after cultured in vitro and treated in different concentrations of ox-LDL for 24 hours.Expressions of eNOS,p-eNOS and caveolin-1(Cav-1)protein were detected by Western blot.The AEC from mice were divided into control group 2 and siRNA group in order to further verify whether ox-LDL attenuates the expressions of eNOS and p-eNOS protein by affecting Cav-1.The expressions of eNOS and p-eNOS protein were detected using the Cav-1 siRNA to knock down the Cav-1 protein.Results The expression levels of Cav-1,eNOS and p-eNOS protein were significantly lower in 25 mg/L ox-LDL group,50 mg/L ox-LDL group and 100 mg/L ox-LDL group than in control group 1(P<0.05,P<0.01)and in siRNA group than in control group 2(0.07±0.02 vs 0.45±0.12,P<0.01;0.34±0.13 vs 0.85±0.38,P<0.05;0.47±0.18 vs 0.86±0.13,P<0.05).Conclusion ox-LDL attenuates the expression of eNOS protein by affecting the expression of Cav-1 protein in AEC from mice.
