摘  要:目的观察真武汤对自发性高血压大鼠血压肾脏功能及TGF-β1/smad信号通路的作用。方法将40只自发性高血压大鼠随机分为模型组、真武汤低剂量组、真武汤中剂量组、真武汤高剂量组,10只正常大鼠做空白对照组。真武汤低、中、高剂量组分别给予含药生药量3.78、7.56、15.12 g·kg^-1·d^-1灌胃,对照组和模型组给予等量生理盐水灌胃,连续给药28 d,分别于0、7、14、21、28 d采用无创血压计检测各组大鼠血压变化。28 d后戊巴比妥钠腹腔注射麻醉,分离血清,剥离肾脏组织。全自动生化分析仪检测大鼠血清肌酐、尿酸、尿素氮水平。酶联免疫法检测各组大鼠血清白细胞介素(IL-6)、肿瘤坏死因子(TNF-α)水平差异。RT-PCR检测各组大鼠肾脏TGF-β1、smad2、smad3、collagen I、α-SMA和CTGF mRNA表达水平。结果与对照组比较,模型组大鼠血压明显升高(P<0.01),血清SCr、BUN、UA水平明显升高(P<0.01),IL-6和TNF-α水平明显升高(P<0.01),肾脏TGF-β1、smad 2、smad 3、collagen I、α-SMA和CTGF mRNA表达水平显著升高。与模型组比较,经真武汤治疗后,各组指标明显改善,以中剂量和高剂量效果较为明显。结论真武汤能够有效降低自发性高血压大鼠收缩压以及炎症反应水平,有效保护肾功能,改善肾脏纤维化水平,其机制可能与抑制TGF-β/smad信号通路,抑制CTGF、α-SMA活化有关。Objective To observe the effect of Zhenwu Decoction on blood pressure,renal function and TGF-β1/smad signaling pathway in spontaneously hypertensive rats.Methods Forty spontaneously hypertensive rats were randomly divided into model group,Zhenwu Decoction low dose group,Zhenwu Decoction middle dose group and Zhenwu Decoction high dose group.Ten normal rats were used as control group.The low,middle and high dose groups of Zhenwu Decoction were given orally 3.78 g·kg^-1·d^-1,7.56 g·kg^-1·d^-1 and 15.12 g·kg^-1·d^-1 respectively.The control and model groups were given the same amount of saline by gavage,the changes of blood pressure in rats of each group were detected by noninvasive sphygmomanometer on days 0,7,14,21 and 28 days,28 days later,pentobarbital sodium was injected intraperitoneally,separation of serum and dissection of kidney tissue,detection of serum creatinine,uric acid and urea nitrogen in rats by automatic biochemical analyzer,detection of serum levels of IL-6 and TNF-αby ELISA,RT-PCR was used to detect the expression levels of TGF-β1,smad2,smad3,collagen I,α-SMA and CTGF mRNA in kidneys.Results Compared with the control group,the blood pressure,serum SCr,BUN and UA levels,IL-6 and TNF-αlevels were significantly higher in model group(P<0.01),and the expressions of TGF-β1,smad2,smad3,collagen I,α-SMA and CTGF mRNA were significantly higher in model group(P<0.01).Compared with model group,after treatment with Zhenwu Decoction,the indexes of each group were significantly improved,with the effect of medium dose and high dose being more obvious.Conclusion Zhenwu Decoction can effectively reduce the systolic blood pressure and inflammatory reaction in spontaneously hypertensive rats,effectively protect renal function and improve the level of renal fibrosis.Its mechanism may be related to the inhibition of TGF-β/smad signaling pathway and the activation of CTGF andα-SMA.
