摘 要:目的观察加味涤痰汤对血管性轻度认知障碍模型(VaMCI)大鼠海马、皮质乙酰胆碱(Ach)、谷氨酸(Glu)含量以及海马神经生长因子β(NGFβ)、脑源性神经营养因子(BDNF)含量的影响。方法本研究采用在大鼠右侧颈总动脉结扎结合半高脂饲料喂食8周的方法,构建血管性轻度认知障碍模型,将大鼠随机分为假手术组、模型组、加味涤痰汤组、尼莫地平组,测定各组大鼠海马、皮质Ach、Glu含量以及海马NGFβ、BDNF含量。结果加味涤痰汤组能够改善模型大鼠的空间参考、工作记忆能力;增加模型大鼠皮质Ach含量,降低皮质Glu含量;提高模型大鼠海马组织NGFβ、BDNF含量。结论加味涤痰汤可通过提高皮质乙酰胆碱水平,调控模型大鼠皮质谷氨酸水平,增加大鼠海马神经营养因子的表达,改善血管性轻度认知功能障碍大鼠的学习记忆能力。Objective To observe the effects of Modified Ditan Decoction on the contents of hippocampus,cortex acetylcholine(Ach),glutamate(Glu),hippocampal nerve growth factorβ(NGFβ)and brain-derived neurotrophic factor(BDNF)in vascular mild cognitive impairment(VaMCI)rats.Methods In this study,rats were ligated with right common carotid artery and fed with semi-high-fat diet for 8 weeks to construct a model of Va MCI.The rats were randomly divided into control group,model group,Modified Ditan Decoction groupand nimodipine ground.The contents of Ach and Glu in hippocampus and cortex,and the contents of NGF and BDNF in hippocampus BDNF were determined.Results The Va MCI group could improve the spatial reference and working memory of rats.The cortical Ach content was increased and the cortical Glu content was decreased.The contents of NGF and BDNF in hippocampus of model rats were increased.Conclusion Modified Ditan Decoction can increase the level of corticoacetylcholine,regulate the level of cortical glutamic acid in model rats,increase the expression of neurotrophic factor in hippocampus of rats,and improve the learning and memory ability of rats with Va MCI.
