新疆南疆某规模牛场牛传染性鼻气管炎PCR鉴定与分析

摘　　要：本试验对15份疑似感染牛传染性鼻气管炎(IBR)患牛鼻腔分泌物样品提取病毒DNA,以gB基因特异性引物进行PCR扩增,测序、核苷酸同源性分析。结果显示,在15个样品中检测出3个牛传染性鼻气管炎病毒(IBRV)核酸阳性样本,检出率为20%,其中1个样本提交GenBank(KY348790),命名为IBRV-4T3-1,3个IBRV核酸阳性样本之间核苷酸同源性达到99%以上,而与IBRV参考株gB基因核苷酸同源性为88.8%以上,构建的gB基因遗传进化树表明,IBRV-4T3-1与参考株亲缘关系较近,处于遗传进化树中同一分支上,属于α疱疹病毒亚科水痘病毒属(Varicellovirus)牛疱疹病毒Ⅰ型(BHV-1)。结合发病情况、临床症状,确认疑似患牛感染IBRV。PCR was used to identify suspected infectious bovine rhinotracheitis in southern Xinjiang. The results can be used as a reference for comprehensive control of IBR. 15 DNA samples of nasal secreta suspension from cattle suspectedly infected with IBR were extracted and detected by PCR with gB gene special primers. 3 of 15 DNA samples were positive of PCR. The amplified fragments were sequenced. The nucleotide sequence of the IBRV-4 T3-1 gene region was deposited in the GenBank sequence database with accession number: KY348790.1. Analysis of the nucleotide sequences and construction of phylogenetic tree were performed based on gB gene of IBRV. The sequencing results showed that the gB gene sequence of IBRV-4 T3-1 had more than 86% similarity to reference strain from GenBank and belonged to bovine alphaherpesvirus 1 of alphaherpesvirus of varicellovirus. The result affirmed that these cattle were infected IBRV.
