摘 要:目的研究玄驹提取物对良性前列腺增生(BPH)的抑制作用及其潜在机制。方法将40只成年雄性SD大鼠随机分成对照组(Control)、模型组(Model)、非那雄胺组(Finasteride)、玄驹组(XJ)以及联用组(XJ&F),每组8只。对照组仅切开阴囊皮肤后缝合,皮下注射植物油作为对照,其余四组行去势手术后,皮下注射丙酸睾酮建立大鼠良性前列腺增生模型,对照组和模型组均给予灌生理盐水,非那雄胺组灌非那雄胺,玄驹组予以灌玄驹提取物,联用组灌非那雄胺加玄驹提取物,持续灌胃4周。4周后取大鼠前列腺组织,称重,计算前列腺指数(PI),通过苏木精和伊红(HE)染色观察前列腺组织病理形态变化,定量聚合酶链反应技术(qPCR)检测前列腺组织中Bax及Bcl-2基因的mRNA表达,并采用蛋白印迹法(WB)检测前列腺组织Bax及Bcl-2基因的蛋白表达。结果治疗4周后,非那雄胺组、玄驹组及联用组大鼠的PI值分别为(1.35±0.15)、(1.50±0.06)、(1.30±0.11),与模型组(1.81±0.15)相比,药物组PI值均明显下降(P<0.05),且前列腺组织增生情况得到了明显改善;非那雄胺组、玄驹组及联用组大鼠的Bax/Bcl-2 mRNA表达比值分别为(1.19±0.72)、(1.00±0.30)、(1.38±0.21),Bax/Bcl-2蛋白表达比值分别为(0.90±0.18)、(0.72±0.13)、(0.92±0.08),而模型组Bax/Bcl-2 mRNA表达与蛋白表达比值分别为(0.81±0.14)、(0.60±0.06),药物组Bax/Bcl-2 mRNA表达及蛋白表达比值均较模型组显著增加(p<0.05),且联用组上调更加明显(P<0.01)。结论玄驹提取物可以有效抑制前列增生情况,并且与非那雄胺联用具有协同增效作用,其作用机制可能与调节前列腺组织内Bax/Bcl-2比例平衡有关。Objective To study the inhibitory effects of Xuanju extract on benign prostatic hyperplasia(BPH)and its underlying mechanism.Methods Fourty male healthy SD rats were randomly divided into the control group(Control),the model group(Model),the finasteride group(Finasteride),the Xuanju group(XJ)and the combination group(XJ&F),and 8 rats each group.The rats in the control group whose scrotum skin was cut and sutured were injected subcutaneously with vegetable oil as a control.After castration,the rats in the other four groups were subcutaneously injected with testosterone propionate to develop the benign prostatic hyperplasia model.The rats in the control group and the model group were given with saline,the rats in the finasteride group with finasteride,the rats in the Xuanju group with Xuanju extract and the rats in the combined group with finasteride and Xuanju extract for four weeks.Four weeks later,the prostate tissues of rats were taken and weighed.The prostate index(PI)was calculated and the pathological changes in the prostate tissues were observed by hematoxylin and eosin(HE)staining.Quantitative polymerase chain reaction(qPCR)and Western blot(WB)were used to detect the Bax and Bcl-2 expressions in prostate tissue.Results After four weeks treatment,the PI values of the finasteride group,the Xuanju group and the combination group were(1.35±0.15),(1.50±0.06)and(1.30±0.11),respectively.Compared with that of the model group(1.81±0.15),the PI values of drug groups were significantly reduced(P<0.05),and the prostate tissue hyperplasia was significantly improved;the Bax/Bcl-2 mRNA expression rations of the finasteride group,the Xuanju group and combination group were(1.19±0.72),(1.00±0.30)and(1.38±0.21),the Bax/Bcl-2 protein expression ratios of the finasteride group,the Xuanju group and the combination group were(0.90±0.18)、(0.72±0.13)and(0.92±0.08),but the Bax/Bcl-2 mRNA expression ratios and protein expression ratios of the model group were(0.81±0.14)and(0.60±0.06),the Bax/Bcl-2 mRNA expression rati
