摘  要:euAP2类转录因子作为一类特殊的AP2/ERF转录因子家族成员,在调节植物开花时间、分生组织活性维持与转化以及花器官属性建成等发育过程中具有重要作用。水稻基因组中共有5个euAP2同源基因,部分成员已被报道参与调控穗分枝、花器官发育、离层发育以及淀粉合成等过程,但是这些euAP2基因在水稻花器官发育调控中功能的协同与分化尚不明确。通过启动子顺式作用元件分析、突变体表型观察以及基因表达模式分析,本研究初步探讨了水稻5个euAP2类转录因子在花器官发育中的作用。结果表明,水稻euAP2家族成员的表达具有特异性,转录水平会因其他成员的缺失而发生改变;euAP2家族突变体的花器官表型不尽相同,仅有OsAP2-3/SNB和OsAP2-5/SHAT1的CRISPR敲除突变体显现明显的花器官异常发育表型,暗示这些基因在调节水稻花器官发育中具有不同的功能。As members of the plant-specific AP2/ERF transcription factor,euAP2 transcription factors play essential roles in plant flowering,meristem activity maintaining and transition,and floral organ specification.In the rice genome,there are five euAP2 homologous genes,which have been reported to participate in the regulating panicle branching,flower organ development,abscission layer development,and starch synthesis.But the synergy and differentiation of their functions in regulating rice flower development are still not clear.Here,we performed promoter cis-acting elements analysis,mutant phenotypes observation,and transcriptional analysis to investigate their specific role in rice flower organ development.Our results showed that the expression of rice euAP2 family members was specific,and the expression level of rice euAP2s was changed when other members were mutated.The flower phenotypes of euAP2 family mutants were different,which were produced by CRISPR/Cas9 technology.Only mutation in OsAP2-3/SNB and OsAP2-5/SHAT1 caused defective floral organ development,suggesting that these genes have different functions in regulating floral organ development in rice.
