摘 要:为了检测不同培养基富集油藏采出液产甲烷体系中微生物群落结构的差异,应用MiSeq高通量测序技术对不同培养基产甲烷体系中细菌和古菌群落结构进行分析。结果表明:原油、GD+S、GD+N和K培养基富集细菌分别检测到181、259、280和305个OTU数,古菌分别检测到72、157、153和132个OTU数。原油和K培养基富集得到的优势菌属较为相似,优势细菌为厌氧杆菌属(Anaerobaculum sp.)和未命名细菌(norank_c_W5);优势古菌为未命名古菌(unclassified_k_norank)、产甲烷囊菌属(Methanoculleus sp.)和未命名TMEG(norank_f_Terrestrial_Miscellaneous_Gp_TMEG_),属于未知功能古菌和氢营养型产甲烷古菌;GD+S和GD+N培养基富集得到的优势菌属较为相似,优势细菌为硫杆状菌属(Tepidiphilus sp.)和厌氧杆菌属(Anaerobaculum sp.);优势古菌为未命名古菌(unclassified_k_norank)和产甲烷囊菌属(Methanoculleus sp.),属于未知功能古菌和氢营养型产甲烷古菌。综合产气量和甲烷含量以及优势细菌与古菌的适配性考虑,最优产甲烷培养基为GD+N培养基。In order to detect the differences of the microbial community structures in the methanogenic system of reservior production liquid enriched by different media,MiSeq high-throughput sequencing technology was used to analyze the community structures of the bacteria and archaea in different methanogenic media.The results show that 181,259,280 and 305 OTU both of which wewe enriched were detected in the bacteria and 72,157,153 and 132 OTU(Operational Taxonomic Units)were detected in the archaea,both of which were enriched in the crude oil,GD+S,GD+N and K media.The dominant genera enriched by the crude oil and K media are similar,the dominant bacteria are Anaerobaculum sp.and unclassified bacteria(norank_c_W5);and the dominant archaea are unclassified archaea(unclassified_k_norank),Methanoculleus sp.and unclassified TMEG(norank_f_Terrestrial_Miscellaneous_Gp_TMEG_),which are mainly unknown functional archaea and hydrogenotrophic methanogenic archaea.The dominant genera enriched by GD+S and GD+N media are similar,the dominant bacteria are Tepidiphilus sp.and Anaerobaculum sp.,and the dominant archaea are unclassified archaea(unclassifie d_k_norank)and Methanoculleus sp.,which are mainly unknown functional archaea and hydrogenotrophic methanogenic archaea.Considering the gas production,methane content and adaptability of the dominant bacteria and archaea,the optimal methanogenic medium is GD+N one.
